Objective: To investigate the expression of HIF-1αHSP90 and phosphorylated Akt(p-Akt) in pancreatic ductal adenocacinoma(PDAC) by high-throughput tissue microarray technique,as well as its relationship with the clinicopathologic features,In addition,the prognostic significance of HIF-1αHSP90 and p-Akt expression in PDAC.Methods: The pancreatic cancer tissue microarray was composed of 88 specimens of the pancreatic adenocarcinoma and 11 normal pancreatic tissues as control subject. Immunohistochemistry was performed to examine expressions of HIF-1α,HSP90 and p-Akt, the relationship of these subjects was analyzed with spearmen rank correlation. In addition,53 Of the 88 cases were follow-up from 2 months to 6 years,in which the survival was analyzed.Results: positive expression of HIF-la,HSP90 and p-Akt in tumors was showed 67(76.1%),65(73.9%),70(79.5%),respectively.The expression of HIF-1αwas closely correlated with tumor size ,TNM stage and lymph node metastasis rather than location of cancer,pathologic grade(P<0.05).The expression of HSP90 and p-Akt was significantly associated with late TNM stage and lymph node metastasis(P<0.01),but no distinct difference was found in tumor location ,size and pathologic grade(P>0.05). Spearmen rank correlation showed a significant positive correlation was observed among this three indexes(P<0.01). Univariate analysis revealed that HIF-1αand p-Akt are prognosticators for PDAC.multivariate analysis showed that only HIF-1αhas independent prognostic significance.Conclusion: Tissue microarray technique is an effective method to detect multiple gene protein expression. The pathogenesis,development and metastasis of pancreatic cancer is associated with HIF-1α,HSP90 and p-Akt .Evently this three proteins play a joint action in development and metastasis of PDAC. THE expression of HIF-1αand p-Akt is a significant prognostic indicator for PDAC.
