Objective:The technique of tissue microarrary (TMA) is used to investigate the expression of pituitary tumor transforming gene (PTTG) among different patients. Then we can analysis PTTG’s role in the development of meningeoma and reveal the identity of PTTG in the cancer genes of human meningeoma. Meanwhile we evaluate the role of JAK/STAT signaling pathway in the occurrence of meningeoma by detecting the expression of JAK2and STAT-5.Methods:1. Sample collection:195samples were collected from the neurosurgery of clinical medical college of Yangzhou University and the neurosurgery of theSecond People’s Hospital of Changshu city fromJanuary2005to December2005and all of them were diagnosed with meningioma.The normal dura tissue were collected from patients with brain trauma decompression operations and which were used as a control group.2. Sample groups:The195meningioma specimens were divided into benign meningioma group, atypical meningioma group and malignant meningioma group according to the assessment of image data, intraoperative fingings and pathological examination. The normal dura tissues were used as the control group.3. The construction of tissue chip:According to the condition of these samples to design microarrays. The microarray of the same patient’s tissue was spotted on the same position by the adjacent two lines (two dots by one case) in order to reduce the system error.4. The accomplish of the slice of immunohistochemistry:The immunohistochemistry Envision method was adopted and the operation was conducted according to the instructions steps seriously. And the tissues ofcolon cancer used as the positive control.5. Statistical analysis:Statistical analyses were performed with SPSS19.0(SPSS Inc.). To assess the relation between different variables and outcome, a chi-square test and rank sum test were calculated with a level of significance P<0.05.Results:1. The positive expression of PTTG in normal human dura mater cell nucleus was showed, while negative expression or mild expression in the cytoplasm. In colon cancer tissues, the cytoplasm showed positive expression while the expression in nucleus is not obvious.2. In the195cases, the positive expression of PTTG in meningioma was172(88.21%). The difference of PTTG expression rates between the different levels of meningioma was statistically significant (P=0.022). There is statistically significant difference in the rates of PTTG positive expression among the different types of meningioma (P=0.0002).3. The expression of JAK2and STAT-5in normal human dura mater tissue cell nucleus was more obvious than the colon cancer tissues, in which the JAK2and STAT5was only obviously expressed in cytoplasm, not in the nucleus.4. There were117positive expression cases (65%) for STAT-5in nucleus from195meningioma cases. The differences of STAT5positive expression between the experimental group and the control group in meningioma was statistical significance (P<0.001). The positive expression cases of JAK2in the cytoplasm was46(23.59%), there is no statistical significance (P=0.05) compared with the control group.Conclusion:1.PTTG and STAT-5were mainly exist in normal brain tissue’s cell nucleus, however, they expressed obviously in the cytoplasm of the meningioma tissues and it’s rate of positive expression was significantly higher than the normal tissue. There was a close relationship between PTTG, STAT-5and meningioma.2. In meningioma, JAK/STAT signaling pathway does not play a major role in activation STAT5, there are at least one other mechanism and some factor can activate STAT-5.3. The advantage of one case two dots technique was that it could maintain the original advantages of the tissue chip, and avoid system error in a better way at the same time.
