Research background Pancreatic cancer is the most common malignant tumors of the digestive system, is one of the main pancreatic duct epithelium from cancer, at home and abroad reported that its incidence trends were significantly increased. Occult pancreatic cancer onset, a high degree of malignancy, early can lead to local tissue infiltration and lymph node metastasis, more than 80% of patients had a clear diagnosis that is in the middle and late, poor prognosis and high mortality rate. To further explore the mechanism of the occurrence and development of pancreatic cancer, to clarify with the tumor cell invasion and metastasis-related molecular mechanisms, diagnosis and treatment of pancreatic cancer is essential.ObjectivesTo explore the heat shock protein 90 (HSP90) and insulin-like growth factor-1 receptor (IGF-1R) expression and biological characteristics of pancreatic cancer among mutual relations in pancreatic cancer. And the invasiveness of pancreatic cancer cells to find and pancreatic cancer invasion and metastasis related to specific factors, in order to find genes related to change and the role of the target reference.Methods (1) 45 cases of pancreatic cancer tissue sect serial sections for 6-8 sheets, respectively, for HE staining and immunohistochemical staining.(2)Using Envision two-step immunohistochemical method detected 45 cases pancreatic cancer and 20 cases of normal tissues adjacent in HSP90 and IGF-1R expression situation.Results HSP90 and IGF-1R expression rate were 73.3%(33/45),62.2%(28/45)respectively in pancreatic cancer, significantly higher than the adjacent normal pancreatic tissue expression of 20.0%(4/20),10.0%(2/20)(P<0.01).Both the degree of organization differentiation were negatively correlated (P<0.01), and lymph node metastasis and clinical stage was positively correlated (P<0.01). both expression were positively correlated (r= 0.670, P<0.01).Conclusions HSP90 and IGF-1R were high expression in pancreatic carcinoma, were significantly different of normal tissue adjacent(P<0.01), both expression were positively correlated (r= 0.670, P<0.01), and play an important role in the occurrence and development process of the pancreatic cancer. Combined detection of HSP90, IGF-1R could be used as pancreatic cancer invasion and metastasis of an objective indicator of the prognosis of pancreatic cancer has important clinical significance.
