Ischemia-reperfusion injury is a kind of patho-process which has been frequent happened in liver disease and wound.It is the one of causes which induces hepatic function injury by surgical intervention in liver transplantation and hepatectomy.The investigation of against ischemia-reperfusion injury in river has became the hot spot of medical domain recently.At present, prevented ischemia-reperfusion injury in liver have three strategies include surgical intervention,medicine and gene therapy. However, many strategies have been used for against ischemia-reperfusion injury in river and it has not been reported thatα-Lipoic acid has been used for against ischemia-reperfusion injury in rat liver.Objective To explore the effect and mechanism ofα-Lipoic acid on ischemia-reperfusion injury in rat river.Methods The models of liver partial ischemia-reperfusion were established in male Wistar rats. The rats were randomized into a control group (group A) and an experimental group (group B) and administered either saline orα-Lipoic acid in 15 min before the ischemia. GPT,GSH,SOD and GSH-Px were examined at various time after reperfusion (1h, 3h, 6h, and 12h) in serum of the rats with or withoutα-Lipoic acid administered. Expression of NF-κB P65 was evaluated by immunohistochemistry assay and expression of MIP-2mRNA and iNOSmRNA was detected by RT-PCR in ischemic liver.Results Compared with group B, the ALT and GSH-Px level in serum, the expression of NF-κB P65,MIP-2mRNA and iNOSmRNA in ischemic liver of the rats in group A were significantly high (P<0.01), while GSH and SOD in serum was lower (P<0.01).Conclusionα-Lipoic acid may protect rats from liver ischemic reperfusion injury through cleaning oxygen free radicals, increasing GSH production, and reducing NF-κB activation and GSH consumption.
