Objective: To investigate the effects of IFNαon the expression of death receptors DR4 and DR5 in human gastric carcinoma SGC-7901 cells and its affection to Trail-induced apoptosis, and approach the possible mechanisms.Methods:The human gastric carcinoma SGC-7901 cell line was incubated with different concentrations of TRAIL for 48h,MTT assay for cell growth and viability. SGC-7901 cell line was incubated with different concentrations of IFNαfor 48h,Semiquantitative RT-PCR to determine the mRNA expression of DR4,DR5,NF-kB,caspase88. The apoptosis rate analyzed by flow cytometer( FCM ).Results: Results show that TRAIL not only inhibits growth and proliferation of SGC-7901 cells, a time- and dose-dependent inhibition, but also induces apoptosis. Semiquantitative RT-PCR analysis shown that IFNαtreatment reduced P75 expression and upregulate DR5,caspase88 expression in SGC-7901 cells. While DR4 was not significantly altered between the groups. The apoptosis rates of SGC-7901 cells were significantly increased in dose-dependent by IFNα.Conclusions: IFNαcan enhance the TRAIL inhibitory effection of proliferation to SGC-7901 cells; IFNαcan up-regulate DR5,caspase88 and down-regulate P75 expression, which is one of the mechanisms of SGC-7901 cells apoptosis.
