Objective:To establish a model of hyperlipemia rat with severe acute pancreatitis.To investigate the influence of hyperlipemia on pathogenetic condition of acute pancreatitis in rats,and the mechanism of lipid peroxidation injury in hyperlipemia rats with severe acute pancreatitis.Methods:Rats received intragastric administration of high fat diet 2 weeks which induced experimental hyperlipemia, and retrograde injection of 3.5% sodium taurocholate into the pancreatic duct which induced severe acute pancreatitis.50 male SD rats were randomly divided into four groups:control group,HL group,AP group,HAP group.After six hours when the models were completed,the value of amylase(AMS),triglyceride(TG),cholesterol(CH) in serum were measured, the changes of malonaldehyde(MDA),superoxide dismutase(SOD),xanthine oxidase(XOD),nitric oxide(NO) in serum and in pancreatic tissue were measured,under the light microscopy,the histopathologic damage degree of pancreas were observed.Results:The serum AMS value in AP group were higher than in control group(p﹤0.01), oedema,hemorrhage and necrosis were detected under the light microscopy;the serum TG,CH in HL group were significantly higher than those in control group(p﹤0.01),but no conspicuous inflammation present in pancreatic tissue;the histopathologic damage of pancreas in HAP group were more serious than in AP group,the level of MDA,XOD in serum and in pancreatic tissue in HAP group were significantly higher than those in AP group,while the level of SOD,NO in serum and in pancreatic tissue in HAP group were significantly lower than those in AP group.Conclusion: A model of hyperlipemia rat with severe acute pancreatitis might be developed by intragastric administration of high fat diet 2 weeks and retrograde injection of 3.5% sodium taurocholate into the pancreatic duct.Hyperlipemia can aggravate the histopathologic damage of severe acute pancreatitis in rats,lipid peroxidation might play a significant role in the mechanism,which hyperlipemia could aggravate the damage of severe acute pancreatitis.
