Objective:The release of inflammatory mediators from mechanical ventilation is one mechanism of ventilator associated lung injury(VALI)and referred to as biotrauma.The intracellular signalling molecules that are activated by ventilation are the same as those utilized by inflammatory cytokines.We investigated the effect of IL-1βon ICAM-1 release from A549 cell and underlying signal transduction pathways.Methods:ELISA and RT-PCR were used to quantify ICAM-1 protein and mRNA expression respectively.Activation of I-kappa B kinase(IKK)was determined by Western blot with specific antibody.Results:IL-1βsignificantly augmented ICAM-1 protein secretion and mRNA expression.IL-1βinduced rapid phosphorylation of I-κB.AS602868, a specific inhibitor for IKK2,abolish phosphorylation of I-κB,and suppressed the expression of both ICAM-1 protein and mRNA.Conclusion:these results suggest that IL-1βinduced ICAM-1 protein secretion and mRNA expression in A549 cells through activation of I-κB kinase.Our data suggests the specific IKK inhibitors could be useful treatments to miniminze the biotrauma.
