Objective In order to study the mechanism of interleukin-1βand leptin on embryo implantation in the early stage, we have observed the expression of intercellular adhesion molecule-1 (ICAM-1) and metalloproteinase 9 (MMP9) intervened separately by different density of interleukin-1β(IL-1β) or leptin which were closely related to the implantation in the cultured human endometrial cells of the mid-secretory phase. We hope to offer the theory to improve the ratio of embryo implantation.Method We got the endometrium in the mid-secretory phase and cultured it. And then we intervened separately by different density of interleukin-1βand leptin. After that we detected the source of the cells and the expressions of ICAM-1 and MMP9 in the endometrium by immunocytochemistry. However, the expressions of ICAM-1 and MMP9 in the medium of model which have been intervened seperately by leptin were detected by enzyme-linked immunosorbent assay (ELISA) or semiquantitative real time polymerase chain reaction(RT-PCR).Results (1) The results of immunocytochemistry showed that all the cells were from the endometrium, which were the endometrial stromal cells and glandular cells. ICAM-1 was mainly expressed in the glandular cells, rarely in the stromal cells, but MMP9 was expressed in the endometrial stromal cells and glandular cells after been intervened separately by different density of by IL-1βor leptin. (2) The results of ELISA showed that that the expression of ICAM-1 in the medium was upregulated after been intervened by IL-1β(lOng/ml) (P<0.05). But after been separately intervened by IL-1β(0.1ng/ml,1ng/ml), they had no significant differences compared with the control group (P>0.05).The expression of MMP9 in the medium was upregulated after been intervened by IL-1β(1ng/ml,10ng/ml) (P<0.05). But after been separately intervened by IL-1β(0.1ng/ml), they had no significant differences compared with the control group (P>0.05).However, the result of ELISA showed that the expression of ICAM-1 in the medium was upregulated after been intervened by leptin (100ng/ml) (P<0.05). But after been separately intervened by leptin (1ng/ml,10ng/ml), they had no significant differences compared with the control group (P>0.05). The expression of MMP9 in the medium was upregulated after been intervened by IL-1β(1ng/ml,10ng/ml,100 ng/ml) (P<0.05). (3) The expressions of ICAM-1 mRNA were increased after been intervened by IL-1β(1ng/ml,10ng/ml) (P<0.05). But after been separately intervened by IL-1β(0.1 ng/ml), they had no significant difference compared with the control group (P>0.05). However, The expressions of MMP9 mRNA were increased after been intervened by IL-1β(0.1ng/ml,1ng/ml,10ng/ml) (P<0.05). The expressions of ICAM-1 and MMP9 mRNA were increased after been intervened by leptin (1ng/ml,10ng/ml,10ng/ml) (P<0.05).Conclusion Different amounts of IL-1βand leptin can separately upregulate the expression of ICAM-1 and MMP9,thus they maybe make full preparation for embryo implantation.
