Studies On Saponins And Pharmacological Activities Of Platycodi Radix

Platycodi Radix is one of traditional medicines and is one of forty general TCM in China, both edible and officinal. It was widely used for opening the inhibited lung-energy, relieving sore-throat, eliminating phlegm, and purulent discharge so on, moreover it is main crude drug. Modern pharmacology indicated that Platycodi Radix possessed many activites such as lowering blood-fat, lowering blood sugar, cut tingcholesterol and anti-obesity, calm and so on. We carried out the studies on platycodins and pharmacological activity of Platycodi Radix to explored extensive plant resources of Jiegeng.The author employed conventional botanical chemical methods, combining P-HPLC to systematically study the components of Platycodi Radix. The author has developed a novel method to simultaneously extract and isolate platycodin D, using HPLC to determine the content of PD in aerial parts of platycodon grandiflorum and to determine its chemical components. Moreover, the author originally did some basic research on platycodins for cytotoxicity and pancreatic lipase activity in vitro, respectively.Twelve compounds were isolated from Platycodi Radix through the conventional chromatography and HPLC isolation method and seven compounds were identified by MS, IR, 1H-NMR, 13C-NMR, 2D-NMR. Among them, a novel triterpenoidal saponin named 16-oxo-platycodin D, together with novel structure, was achieved, and the other six known compounds are platycodin-D, deapioplatycodin-D, platycoside-E, deapoi-platycoside-E, platycoside F, polygalancin D, respectively.PD was not only the index of quality control for platycodi Radix, but also is the main saponin in Platycodin Radix, and its related products and no reports on selling PD have appeared in papers in China. The author has developed a new method to simultaneously isolate PD by doing a number of experiment, with its degree of purity≥98.8%(HPLC), and total 4.2g. Now PD was provided with around twelve research institutes in China as control article.Cytotoxic reaction showed that PD can effectively inhibit the growth of cell Bel 7402, BGC823, MCF7, respectively. Its half killing degree was 11.70μg/mL,9.10μg/mL,17.72μg/mL, respectively, while PD₃ and polygalacin D had little inhibition on the growth of cell Bel 7402, BGC823, MCF7, respectively.Meanwhile, the content of PD in aerial parts of platycodon Radix was originally determined, building up the foundation for further exploring the aerial parts of platycodi Radix and enlarging the sources of Chinese Traditional Medicine.
