| Objective:Isatidis Radix as one of staple herb-medicines in Gansu Province,it has important pharmacological activities.Isatidis Radix has appeared thousand years in our life,however,in recent years,the quality of traditional Chinese medicine(TCM)had downslide which lead to degeneration.The reason for this phenomenon is that people pursue the personal interest,use pesticides and plant growth promoter and destroy the plant growth characteristics.For these reasons,the research colleted Isatidis Radix from different place and planted in hezheng,finally study on determination of effective components and pharmacological activity for different ecotype from Isatidis Radix,ultimately select the fine varieties.Methods:1)The microscopic characteristics of 11 specimens Isatidis Radix were described.According to Chinese Pharmacopoeia,the moisture,total ash,acid-insoluble ash,ethanol-extract were tested;The polysaccharide from Isatidis Radix was determined by phenol-sulphoacid;The contents of guanosine,uridine,epigoitrin,adenosine,benzene acid,salicylic acid,indigo,indirubin were measureed by HPLC.2)Using HPLC and “Similarity Evaluation System for Chromatographic Fingerprint of TCM”,the Isatidis Radix finger print of CHCl3 part was maked on the basis of chemical constituents(epigoitrin,benzene acid,salicylic acid,indigo,indirubin).3)Using total ash,ethanol extract,polysaccharide,guanosine,guanosine,uridine,epigoitrin,adenosine,benzene acid,salicylic acid,indigo,indirubin as evaluation index,using three mathematical statistics methods and SPSS 19.0 to evaluation and sorting the quality of Isatidis Radix.4)Reference the result of ranking results,choose some samples to implement the pharmacological experiments.And using the auricular swelling ?paw edema and ga-sbag synovitis model to observe the anti-inflammatory effects of Isatidis Radix,the immunosuppressed model were established by injection of cyclophosphamide(CTX)to study the effects of Isatidis Radix on index of thymus,bloodroutine,cytokine.Result:1)Through the microscope observed ducts?starch granules?caudexs?stone cells structure.The single grain is abundant in starch granules and the form isspherical,oval or hemispherical,also has compound starch;The water content,total ash,acid-insoluble ash,ethanol-extract,polysaccharide,guanosine,uridine,epigoitrin,adenosine,benzene acid,salicylic acid,indigo,indirubin were 7.34%~8.75%,4.02%~ 5.70%,0.39%~ 1.07%,35.32%~ 42.66%,9.43%~ 17.57%,0.23mg·g-1~ 0.36 mg·g-1,0.04 mg·g-1~0.08 mg·g-1,1.57 mg·g-1~3.15 mg·g-1,0.32 mg·g-1~0.62 mg·g-1,3.36 ?g·g-1~10.07 ?g·g-1,5.51 ?g·g-1~19.62 ?g·g-1,0.98 ?g·g-1~15.43 ?g·g-1,1.21 ?g·g-1~10.58 ?g·g-1.2)“Similarity Evaluation System for Chromatographic Fingerprint of TCM”was used to analyze similar degree of 11 specimens Isatidis Radix,the result was that between sample and sample similar degree is 0.600~0.995,the most sample similar degree are closed(greater than 0.900)except BLG2.3)Using grey relational analysis,principal component analysis,cluster analysis to evaluate the quality of Isatidis Radix,although each method had not same result,but the final result was that BLG2 were better.4)Different ecotype of Isatidis Radix could reduce the swelling of the mice auricle,paw edema and total protein,LTB4 and MDA in airbag synovitis exudates,and upgrade serum levels of superoxide dismutase(SOD);Degrade the tumor necrosis factor-?(TNF-?)and upgrade the index of thymus,the number of red and white corpuscles,the level of interferon-?(IFN-?),interleukin-4(IL-4)(P<0.05,0.01)of mice immunosuppressed model;Above the research of anti-inflammatory and immunomodulation,there are no significant differences between Isatidis Radix of Gansu different strains and tetraploid.Conclusion:The contents of each chemical constituents is differencent,although tetraploid Radix and diploid Radix have some differences in CHCl3 fingerprints and quality evaluation,but there are no significant differences between each sample in anti-inflammatory and immunomodulation,provide basis for the use of Radix. |
