To utilize deeply and effectively the source of Platycodon gradiflorum (Jacq.) A. DC and develop new medicines, researches were carried out systematically for preparation of total saponins and for preparation, separation, qualitative and quantitative analysis of prosapogenins from Radix Platycodi, and the bacteriostastic activities were investigated.ZTC-1 microreticular resin was used firstly in the technics to extract and isolate platycodins with water and 20%. 30%. 40%. 50%. 95% ethanol as eluant. The method was simple and had an average gaining rate of 2.05%. However it needed a long period. Therefore another method was established subsequently for preparing platycodins by water-extracting and macroreticular resin column chromatography-separating. i.e.. to the powder of Radix platycodi, added altogether 20-fold water and refluxed three times with 0.5 h for each time. The filtrate was then passed through the macroreticular resin and platycodins obtained by gradient eluation using water and alcohol as eluant. The method offered a basis for industrialized production of platycodins for its high average gaining rate, 1.603 %. easy operation and low cost.3 % potassium hydroxide solution of platycodins was acidified to pH = 4 after refluxed in mildly boiling water for 0.5 h. and was extracted three times with the mixture of methanol and n-butanol. The organic phase was combined and concentrated to give prosapogenins. The average production rate was 63.38 %.
