The Effect Of Douching By Hydrogen Peroxide To Brain Tissue

Objective: TO study the effect of douching by hydrogen peroxide to brain tissue. To observe the changes of the brain tissue douched by hydrogen peroxide, the effect of hydrogen peroxide to the tissue and blood vessels, and through the expression of glucocorticoid receptor in the tissue, and the concentration of glucocorticoid in blood, to analysis the reaction initially.Methods: The hydrogen peroxide douched models were established by rats. All rats were distributed randomly into six groups: every group have 14 rats, and every group take suction randomly 4 rats as control group. Rats were douched by 3%hydrogen peroxide ,at the 1st, 6th, 12th, 24th, 48th, 72nd hour after douched, all rats were killed. The control groups were same state as others except douched by NS. Expression of GR was detected with immunohistochemical staining and MDA, SOD were quantified with spectrophotometer assay. And use pathologic methods, HE staining to detect pathologic changes of the tissue and the blood vessel.Results: Compared with control group, the MDA content, the SOD activity, the expression of GR, and the concentration of glucocorticoidin blood were significantly different. The results were 13.6817±0.3591(P< 0.05), 13.0817±0.4332(P < 0.05), 14.5729±1.0402 ( P < 0.05 ) , 11.3274±0.2438(P<0.05) respectively. Through morphologic observe we found in the tissue which douched by hydrogen peroxide, the pericaryon were necrotic, the caryon appeared karyopycnosis, quassation, spallation and the bioblasts were necrotic too. The diastem around cells were enlarged, the tissue changed more loose,and we also found there were a few punctuate hemorrhagic focus. Some of the blade of the tissue which douched by hydrogen peroxide, we found vascular endothelial cell appeared reductus, even there some VEC were damaged, the smooth muscle cells were twisted. The ectal membrane changed more thick, and inflammatory cell infiltrate.Conclusion: The tissue happened fervent lipid peroxidation after douched by hydrogen peroxide. Neurocyte and blood vessel were damaged. The expression of glucocorticoid receptor was decreased, and the concentration of glucocorticoid was elevated.
