Rhizoma Chuanxiong and Rhizoma cnidium are traditional medicines in China and Japan, respectively, both of which are belonged to one of traditional medicine which are called "xiong qiong". In addition, Ligusticum chuanxiong Hort. is included in CAP(2005 edition),and it have been wildly used for treatment. On the other hand, Cnidium officinale Makino is included in Pharmacopoeia Japonica officially, and it has been used in most of preparation of traditional medicine in Japan.In order to study the relationship between L. chuanxiong Hort. and C. officinale Makino, provide scientific evidences for their classification and identification. A systematic pharmacognostic comparison and RAPD and ISSR anaylsis about them is studied in this paper.1. It has identified the plant, appearance and microscope character. The results showed that the L. chuanxiong Hort. differ from the C. officinale Makino in the leaf, stem and the mode of proliferate significantly. Meanwhile, the degree of size in the transverse section of stem skin along with distribution of oil cell has differences between L. chuanxiong Hort and C. officinale Makino.2. Applied technology of Random Amplified Polymorphic DNA (RAPD) first time, it has Amplified 35 samples which come from 2 area by adopting 15 selected primers and it has applied clustering analysis. And then the analysis of cluster has carried out. 3. Applied technology of Inter Simple Sequence Repeat (ISSR) first time, it has Amplified 20 samples which come from 2 area by adopting 4 selected primers and it has applied clustering analysis. And then the analysis of cluster has carried out.4. It has isolated 8 compounds from the extractive of radix chuanxiong, and identified these by the way of TLC, HPLC and ~1H-NMR. They are palmitic acid, levistolide A,β-sitosterol, pregnenolone, daucosterol, ferulic acid, protocatechic acid, Caffeic acid.5. RP-HPLC method is used for studying the contents extracted by ethanol. The result indicates that the number of peaks between two plants is not obviously different, but area of peaks have great different.Thereinto, 1#,4#and 5# peaks could be looked as diagnostic peak.
