Arsenic is an element which is widespread distributed in the natural world, its Clarke is 5×10-4 and cosmic abundance is 4.0. In addition to some natural arsenic, there has been about 150 kinds of arsenious mineral. It is reported that some Traditional Chinese Medcine, like arsenolite ore, realgar, arsenic trisulfide, contains arsenic, and also some frequently used Traditional Chinese Medcine contains arsenic. The biological effect of arsenic correlates with its dosage and speciation. For example, the half lethal dose (LD50) to mouse of arsenic trioxide is 20mg/kg, and that of dimethylarsenic is 700-2600 mg/kg, that of arsensugar is more than 8000mg/kg.Chinese crude drug is widespread, and its place of production, collection, processing drugs affect arsenic's content. It is very important to test the content of arsenic to insure its safety and standard.Capsule of YUBAO, which is triturated by our institution, has been used in Xijing Hospital of The Fourth Military Medical University. It is convenient to use and its curative effect is reliable with little side effect. But its arsenic content was tested high in lab frequently. So in this study,we will investigate arsenic content and formation in defferent kinds of TCM and Chinese formulated products. It is very important to investigate the effect of arsenic's biological dose-effect relationship on genital or hematopoietic system.PurposeTo study the possible mechanism of action of CYB to treat male infertility and investigate the arsenic's dose-effect relationship between genital system and hematopoietic system by test the arsenic content of the group.Method1. To analyse arsenic and its speciations in CYB.1.1 Physical and chemical identification and preparation technology of CYB: Physical and chemical identified for the TCM to ensure its quality and amplified according to modo praescripto.1.2 Arsenic content and speciation in CYB and its active compound: Use AFS to analyse arsenic content in CYB. The high arsenic content substances are been Choosed and analysed the arsenic speciations.1.3 Uncertainty study for the arsenic content TCM: To discuss the uncertainty influence factors through arsenic content alalyse progress by AFS by use of direct cause and effect graph to find out the main influencing factors.2. Method of experimental rat model mockupRats were exposed in Co60, than we find out the suitable expose dose for male rats'reproduction functional disturbance and hematopoietic system inhibition.After irradiation 3 months, the number of spermatozoa, abnormity rate, persistence rate and self-recovery phase were observed. 3. The biological dose-effect relationship of arrhenic3.1 The effect of YBC and different dosage As2O3 for rats'genital and hematopoietic system: The rats were exposed in Co60 and the model was build. The rats were divided in to different groups according to different dose of YBC (0.2,0.15,0.1 g/mL),different dose of As2O3( 0.15,0.3,0.75mg/mL)and model group, actinolitum group,control group.Different groups were treated differently.Three weeks later, blood were got and the rats were put to death humanitarianism. Dissect those animals and observe the index of the genital system. Compare the main organ coefficient and body weight growth factor, Bone marrow of sternum slides are common dyeing observed.3.2 The arsenic content of arsenic administration team rats'tissue: AFS is carried out to analyse arsenic content in the medication administration team's main organ and tissue to find the imaginabale arsenic cumulative organ or target organ.Results1. Arsenic content and speciation in YBC and its active compound: The high arsenic content in Yubao in caused by coptis chinensis, actinolite, oyster, keel. It is mainly trivalence inorganic arsenic in YBC. We established the mathematic model via on the uncertainty study through arsenic analysis by AFS. Each constituent of the uncertainty were subdivision hypothesis, and each results were discussioned.2.Screening the method of experimental rat model mockup:The differences of spermatozoa number, persistence, abnormity among each group which was exposed in 4.0Gy, 5.0Gyad to the normal group were significant(P<0.01). The differences of spermatozoa number, persistence between groups which was exposed in 4.0Gy, 5.0Gyad were significant(P<0.05). There was no significant abnormity difference between the groups. None was lead to death throughout the progress by complication.3.The biological dose-effect relationship of arrhenic:There were significant difference between the model group and normal group in spermatozoa number, persistence, abnormity (P<0.01),that is to say the rats model was stable and reliable. Compared with the model group, the CYB groups and low arsenic intaked group were higher in the number of spermatozoa(P<0.05).Compared with the model group, the CYB groups and low As2O3 intaked group were higher in the number of spermatozoa(P<0.05), the Yubao groups and low As2O3 intaked group were lower in teratospermia rate (P<0.05). The LH,T, E level in the model groups were lower than that in normal group, there was significant difference in T and E level(P<0.05). Compared with the model group, the E level in As2O3 low-dose group and T were higher (P<0.05), and also the T level of YBC groups was higher, the media-dose YBC group was significant.Compared with the normal group, there was significant difference in epididymis coefficient, testicle coefficient, no significance in spermary coefficient, prostate coefficient, cryptae odoriferae coefficient to the model group. Compared with the model group, testicle coefficient in As2O3 high dose group was lower, however, higher in Yubao low and media dose group(P<0.05).There was no significant difference in body weight coefficient between the normal group and the model group, more higher in As2O3 high dose group, lower in actinolitum group. (P<0.05).There were significant difference adrenal coefficient between the normal group and model group, As2O3 high dose group (P<0.05).No significance in thymus coefficient, spleen coefficient and kidney coefficient. No significance in cardiac coefficient, liver coefficient and pulmonary coefficient between model group and normal group. There was significance in cardiac coefficient, liver coefficient and pulmonary coefficient between As2O3 high dose group and normal group (P<0.05). Spleen coefficient in As2O3 low dose group was lower than that in normal group (P<0.05).Bone marrow hyperplasia was active in normal rats, there was no change in form of bone marrow hyperplasia, granular cell, achroacyte.Being exposed in Go60 marrow apomorphosis widely,megalokaryocyte,giant plates,leukomonocyte, erythrolysis, bodies, cell membrane unsharpness,cell morphoge changed,Cellular proliferation to the utmost, vacuolar degeneration,much lattice fibers.There was vacuolar degeneration in arsenic high dose group, actinolitum group.It's more than model group but also difference compared with normal group.Marrow aspect anastate better in low arsenic dose group and medium dose group,YBC intake group than that in other groups.Conclusion1. Those medical materials are rich of arsenic: coptidis rhizome: 188mg/kg; actinolitum: 115mg/kg;oyster: 22mg/kg;keel: 41mg/kg.The main arsenic speciation in TBC was As(Ⅲ).2. The bone marrow depression and infertility model rats can be established by use of 5.0GyCo60 exposion.3. Therapeutic dose of CYB can improve the spermatozoa number, abnormity rate, persistence rate, and also enhance the T level postdose, while not in estrin level. Actinolitum in CYB could not enhance spermatozoa quantity and hormone level, to hint that Yubao's therapeutic action for sterility rats dose not depend on its arsenic content.4. High dose As2O3 could accelerate the body weight gain of the anaplasis rats. Actinolitum could cause the deficiency of kidney-YANG deficiency medel.5. Adrenal coefficient in model group and high dose As2O3 group was higher than that in normal group; High dose As2O3 could significantly enhance the rats'cardia, liver pulmonary coefficient.6. CYB, medium, low dose As2O3 can recover bone marrow of sternum condition of the ionising radiate impaired rats.7. Low dose As2O3 was ingested by rats coud be discarded by via metabolic mechanism and not be cumulated. But with the increase of arsenical dose, it's cumulated quickly, arsenic content quickly increase significantly. While it is uneven distributed in the tissue, High in fur, liver, spleen, lung, kidney.
