【Purpose】To investigate the expression of cycloxygenase-2 in the human pterygium fibroblasts(HPF)and expression of cycloxygenase-2 also in the human pterygium fibroblasts following vascular endothelial growth factor (VEGF) and basic fibroblast growth factor (bFGF) treatment.【Methods】The positive expression of cycloxygenase-2 in 30 HPF was determined by the immunohistochemistry and the control group 30 normal conjunctival organization. Cell culture experiment was performed using cutted human pterygium. The cultured 3 or 6 passage of human pterygium fibroblasts (HPF) were used in the next steps.HPF were determined by the method of SABC. Expression of cycloxygenase-2 was detected by Western blot in HPF following vascular endothelial growth factor (VEGF) treatment at different concentrations(1﹑ 10﹑ 100ng/ml). Expression of cycloxygenase-2 was detected by Western blot in HPF following basic fibroblast growth factor (bFGF) treatment at different concentrations(1﹑10﹑100ng/ml).【Results】The immunohistochemistry test demonstrated that the positive expression of COX-2 in HPF was 73.3%. In nomal conjunctiva organizations positive expression rate is 0 (P<0.01). Positive and negative vimentin expression were detected keratin by the method of SABC. The expression of cycloxygenase-2 increased in culture HPF treated with VEGF at different concentrations. The expression of cycloxygenase-2 increased in HPF cultures treated with bFGF at different concentrations.【Conclusion】This study showed a positive expression of COX-2 in HPF. Both VEGF and bFGF are dosely induced the expression of COX-2 in the HPF. COX-2 may play an important role in the process and development of pterygium.
