Partâ… The Expression of Leptin and Its Receptor in Cervical Cancer and Its Relation to Angiogenesis and Tumor GrowthObjective The purpose of this study was to evaluate leptin and leptin receptor (OB-R) expression in the cervical cancer and their correlation with Angiogenesis and clinicopathological parameters.Methods1. 10 cases of chronic cervicitis,12 cases of cervical intraepithelial neoplasia and 50 cases of cervical cancer were examined by immunohistochemical studies with anti-leptin and anti-OB-R.2. Microvessels were immunohistochemical labled with an antibody to CD34. Microscope analysis was used to evaluate microvessel density (MVD).Results1. It was only in stratified squamous epithelium of chronic cervisitis and cervical intraepithelial neoplasia that cells were visualized by staining with leptin antibody or anti-OB-R; Thirty-nine (78%) and thirty-three tumors (66%) were classified as leptin and OB-R positive respectively.2. Leptin expression, MVD significantly correlated with degree of differentiations, invasive depth and distant metastasis (P <0.05), conversely, did not correlate with age and histological types.3. At the same time, OB-R expression was associated with invasive depth and distant metastasis, however, it was independent on age, histological types and degree of differentiations.4. Tumors expressing leptin or OB-R had a significantly higher MVD than those non-expressing leptin or OB-R. In addition, there were no difference between leptin positive and OB-R positive.Conclusions Our results suggest that local overexpression of leptin and its receptor in cervical cancer may contribute to tumor progression by increasing angiogenesis and tumor growth.Partâ…¡The Clinical Value of Expression of Leptin in Exfoliated Cells for Screening High-risk Group of Cervical CancerObjective To search a new method for screening high-risk group of cervical cancer.Methods 13 cases of chronic cervicitis, 46 cases of dysplasia of cervical epithelium, 15 cases of carcinoma in situ and 20 cases of squamous cell carcinoma were collected. The positive expression of leptin in these tissue were tested by immunohistochemical study labled with antibody to leptin, and that in these exfoliated cells samples were examed by immunocytochemical technology respectively.Results1. With the progress of cervical lesions (from chronic cervicitis to squamous cell carcinoma), the positive rates of leptin expression in cervical histopathotogical paraffin slices were gradually increased, so did the leptin expression in exfoliated cell samples and the two increased trends had obvious relativity (P<0.05).2. The leptin expression in paraffin slices of chronic cervicitis had no difference compared with dysplasia of cervical epithelium (P>0.05), while had significant difference compared with carcinoma in situ or squamous cell carcinoma (P<0.05); at the same time, there were significant difference among severe dysplasia , carcinoma in situ and squamous cell carcinoma(P>0.05).3. There were no difference of leptin protein expressions in exfoliated cells samples between moderate dysplasia of cervical epithelium and chronic cervicitis(P<0.05), while the positive rates of leptin expression in severe dysplasia , carcinoma in situ and squamous cell carcinoma were higher than chronic cervicitis respectively(P<0.05). In addition, there was no difference among those three groups.Conclusions Our results suggested that leptin expression in exfoliated cells were associated with the progress of cervical lesions, so it may serve as a new simple method for screening high-risk group of cervical cancer and leptin may be a promising biomarker of cervical cancer.
