Danggui Blood-Supplementing Decoction is created by Li Dong-yan of the Jin-Yuan Dynasties. This herbal prescription is made up of Radix Astragali and Radix Angelicae Sinesis. This decoction is used to treat various clinical ailments and diseases as it benefits energy and nourish blood in our body system. In recent years, it is observed that majority of the research reports were mainly focused in the area of medical studies. However, the relationship between chemistry studies and clinical therapy are less researched on. This article, based on the foundation built from international survey, shows a series of researches on the chemical composition of Danggui Blo0d-Supplementing Decoction. It established a new method for the determination of trace dements, ferulic acid and carbohydrate compounds in Danggui Blood-Supplementing Decoction. It also investigated the differences between the solubility of the chemical compositions. The different ratio proportions and extraction method of Danggui Blood-Supplementing Decoction were compared for the trace elements, ferulic acid and carbohydrate compounds. The main researches as follow:1. A new method for the determination of selenium in Danggui Blood-Supplementing decoction by atomic fluorescence spectrometry is proposed. Comparison was made between the contents and solubility of selenium in different decoction proportions and extraction methods. In the different decoction proportions, the effect of phospholipids on the solubility of selenium was investigated. The average recovery of selenium is in the range of 83.82%~95.04%. The solubility of selenium is higher in the ratio proportion of 1:5 (Radix Angelicae Sinesis: Radix Astragali, w/w), as compared to ratio proportion of 1:1. The method proved to be simple, rapid, sensitive and accurate.2. A method for the determination of Pb, Cr, Cd, As in Danggui Blood-Supplementing decoction by inductively coupled plasma atomic emission spectrometry is proposed. Comparison was made between the contents and solubility of these heavy metal elements in different decoction proportions and extraction method. This experiment demonstrates that Radix Angelicae Sinesis decoction, Radix Astragali decoction and Danggui Blood-Supplementing are completely digested by nitric acid. This satisfies he requirements of the ICP-AES detector. The average recovery of this method for these heavy metal elements fall in the range of 83.0%~96.0%. The RSD is in the range of 0.36%~4.67%. In addition, the contents of heavy metal in the solution of ratio factor of 1:5 (Radix Angelicae Sinesis: Radix Astragali) are lower as compared to the ratio factor of 1:1. This result has backed up the rationale behind the classic formulation of Danggui Blood-Supplementing Decoction, as well as the s acknowledged beneficial clinical therapeutic effects. This research has also shown the effect of phospholipids compounds in Jiangxi black-bone silky fowl on the solubility of these heavy metal elements in different proportions of Danggui Blood-Supplementing Decoction. This aid develops the medicinal value of Danggui Blood-Supplementing Decoction appropriately,3. The analysis on the content of ferulic acid in Radix Angelicae Sinesis Blood-Supplementing decoction is carried out by using an anion analytical column, a column including IonPac AS18 column (2*250mm) arid protective column, IonPac AG18 (2*50mm). The solvent used is 0.055mol/L NaOH. Studies are carried out on the chromatography separation conditions of the ferulic acid and other coexisting factors. A new method for the determination of ferulic acid was established. The method used to detect the content of ferulic acid was limited to 0.2142mg/L, with a resolution of 0.9999 and average recovery of 96.6%~104.9%. The outcome gave a satisfactory result for the content of ferulic acid in Danggui Blood-Supplementing Decoction.4. A new method has been established to detect the content of Arabinose, Glucose, Fruotose, Riboss and Sucrose in Danggui Blood-Supplementing Decoction by using anion-exchange chromatography-integrated pulsed amperometric detection. The chosen chromatogram conditions are as followed: Carbopac PA10 anion analytical column is used as the chromatography column, 0.055mol/L NaOH as the solvent with a flow rate of 0.18mL/min. Studies has been carried out on the chromatogram separation condition of the carbohydrate. Under the optimized condition of the separation analysis, the peak area and resolution of the content of the various monosaccharides and Sucrose give good results. The limits of detection (LOD) of the various sugars are in the range of 0.0038~0.2005 mg/L, with an average recovery that falls in the range of 89.37%~97.80%. This method is easy and convenient to be carried out. At the same time, the differences in the solubility of carbohydrate in different Danggui Blood-Supplementing Decoction, using different extraction method is investigated.
