Amaranthus retroflexus L , which belongs to Amaranthus family, is Annual herbage. It grows in America originally, and then spread abroad all parts of the world. In our country ,it mainly distributes in the Northeast, North of China and every provinces along the Yangtze River valley. It grows in the farmland, aside the ground, around the residence and so on, and usually regarded as weed by the peasant, mostly become harmful to many kinds of crops on land such as cotton, beans, peanut, melon, potatoes, vegetables etc. Its tender leaves and stems are eximious edible vegetables and also fine pig feed. The whole grass and seed of Amaranthus retroflexus L can be used as medicine, which can treat enteritis, dysentery, abscess of throat, sore, haemorrhoids disease of swelling and aching etc, has the function of clearing away heat on detoxifying, clearing liver and making eyes bright, cooling blood and dispelling turgescence. According to foreign reported in the literature, its whole grass can be used in adiposity, chronic uterus inflammation and goitre in clinic.Though Amaranthus retroflexus L has commonly used as a Chinese herbal medicine in some of China's regional liber, but research materials on its chemical components, pharmacology and clinical research are few at present, and its research depth is far from enough. In order to explore the valuable experience of folk traditional Chinese medicine, prepare to the further study and development for this officinal resource , this thesis carries on the research on the following several respects: 1. We summarized the current situation of Chinese traditional medicine treatment on hypertension, and the chemical composition, pharmacology activity and clinical research of the Amaranthus. Which offer valuable reference for the research of Amaranthus retroflexus L.2.Using the method of organic solvent extraction, column chromatography, extraction, re-crystallization, we obtained 10 compounds from Amaranthus retrofleexus L. with their physical and chemical properties, through UV, IR, MS, 1H-NMR, 13C-NMR , HMBC, HMQC spectra techneque, we accomplished eight compounds'structure identification in all. They are: L-Tryptophan(I), Rutin(II),α-spinasterol(III),β-sitosterol(IV),kaempferol-3-O-rutinoside(VI),stigmasterol(VII),n-triaconatanoic acid(IX)。Compound I and Compound VI were the first isolated from Amaranthus retroflexus L.3. The volatile oil from Amaranthus retroflexus L was investigated with the GC-MS. We identified 26 compounds thereinto. with the peak area normalization method, we calculated the relative content about those component, and showed the published component accounts for 78.2% of the total amount of the whole oil.4. We gave pharmacology activity research on Amaranthus retroflexus L. It showed that the EtOAc exract of Amaranthus retroflexus L could prominent depressed those big mouse's blood pressure. Because this components contain a large number of flavone compound, we conjectured flavone compound are main active ingredients which play the antihypertensive function in principle.5. we got the total flavone′s content of Amaranthus retroflexus L by colorimetry。The experiment result shows that its total flavone content is approximately 2.06mg/g on average. The method is easy, requirement for apparatus is little, the linear relation is good. So we set up a relative suitable determine method for the total flavone in Amaranthus retroflexus L,which not only provide a science gist for the further depth exploitation of Amaranthus retroflexus L ,but also play an active role in looking for new natural flavone compound resource, improve the utilization ratio of plant resources, protect the natural resources.
