The alcoholism may cause many internal organs like livers, the gallbladder, the cardiac muscle harm, the alcohol also may cause the intelligence and the memory drops, and appear various psychiatric symptom. Some research indicated the alcoholism may cause the hyperlipemia, the hypertension, and causes the change of erythrocyte deformability, the platelet aggregation abnormality, to reduce the brain regional blood flow, induce arhythmia, increaseβ-endogenous opioid peptide composition, to cause the ion disorder and so on, more -over these aspects are dangerous factors to urge stroke. The alcohol-olism causes the brain damage mainly is spot nerve cell diminutions in number and the varying degree cell degeneration in cerebral cortex and hippocampus. And also may causes the number of nerve cell, the granular cell and Purkinje cell in cerebellar cortex decrease and the cell degeneration. In alcoholism rats fold in the endangium of small vessel and some endothelium exfoliation were found. Because after the alcoholism the cerebral vessels may appear above a series of changes, regarding with blood vessel close correlation vascular endothelial growth factors whether also can appear the corresponding change not to have the further research. Therefore this experiment has prepared the rat alcoholism animal model, carries on the observation through the light microscope to the rat cerebral cortex and the hippocamp pathology change, and the application immunity class method carries on the rat cerebral cortex and the hippocamp VEGF determination, and has carried on the discussion to the VEGF change mechanism. Applies clinically in on whether to VEGF treats the cerebrovascular disease which the alcoholism causes to play the corresponding role to discuss.Objective: To observe the pathology change and change of VEGF in cerebral cortex and hippocampus in alcoholism rats and discusses the related mechanism.Method: After the Wistar maleness rat feeds 1 week to start normally to make the mold. Alcoholic group:Every rat was fed with 50% alcohol two times everyday according to 8ml/Kg,the interval of time is 6 hours. Two weeks later, every rat was fed according to 10ml/Kg. A week later they were fed according to 12ml/Kg,the intervals of time are all 6 hours. The total time of this test is four weeks. Control group : every rat was fed with the same dosage of 0.9% sodium chloride at the same time, and the total experimental time is four weeks, too. During the experiment we observe the two group of rats with their general condition, measure their weight periodically. Two groups of rats intragastric administration 4 weeks after the 6%chloral hydrate abdominal cavity injection anaesthesia, opens the chest cavity, in turn application physiological saline, the paraformaldehyde pours into through the ascending aorta and takes out the brain organization, after further fixed dehydrates, the embedding, the histo-pathology uses the convention tissue slice, HE dyes and carries on the light microscope observation brain organization pathology change, and determines VEGF with the SP law.Result: After 4 weeks there are 17 rats survived in the alcoholic group, the mortality rate are 29.2%,and there are 15 rats survived in the control group , in which one death because of operation mistake. After 1 weeks the control group rats can adapt intragastric administration operation, the body weight normal growth, but the experimental group rats always exists intragastric administration to be difficult. After 2 weeks they appear the hair withered and feed quantity reduction disorderly. After 3 weeks the experimental group rats appears the hair to fall off, tiredly, the malnutrition, becomes emaciated, moreover some also appears the performance which the nervous system damages, for example: hemiplegia. And two group of rats' body weight (g) differences have statistics significance (P<0.01); The alcoholic group rats hippocampus cell arrangement is irregular, the level is chaotic, pyramidal cell diminution in number, some appearance cell slight denaturation, atrophy. Cerebral cortex spongiocyte proliferation, some appearance colloid bouton and phlogocyte infiltration; The control group rats cerebral cortex and hippocampus VEGF have not expressed, Alcoholic group rats cerebral cortex 5 rats VEGF is the strong masculine expression, 7 rats are the masculine expressions, 3 rats for the weak masculine expression, 2 rats have not expressed. Hippocampus area 7 rats for the strong masculine expression, 6 rats are the masculine expressions, 2 rats for the weak masculine expression, 2 rats have not expressed.Discussion: The cerebrovascular disease pathological change and the corresponding brain organization damage in the alcoholism experimental animal model is requested to be similar to change of the brain organization in the humanity alcoholism. The effective model must be the pathological change has certain developing process, the speed of formation is high, the mortality rate is low, makes the norm law easyly to do, and after makes the mold to stop the pathological change reversing slowly, or forms the irreversibility change to be advantageous for conducts the medicine intervention research. This experiment through carried on to the rats intragastric administration the method to prepare the alcoholism, could endure in the rat under the condition enlarged gradually intragastric administration the alcohol dosage, guaranteed intragastric administration frequency, after tested 4 weeks the rats appear encephalopathic performance: hemiplegia. This way may guarantee makes the mold indeed qualitatively, and can avoid enlarging the dosage to cause the animal death blindly, simultaneously may save the run time. This experimental study discovery alcoholism brain tissue can appear ischemic and anoxemia change and the brain damage.
