Background:Periodontitis is a periodontal support organizations that do not reversible loss of bacterial infections as a result of the loss of teeth,chewing the main reason for the loss of function.The highest goal of periodontal treatment of periodontal tissue defects is to repair and restore normal periodontal tissue structure and function.The key of periodontal tissue regeneration is the regeneration of alveolar bone and cementum,the current treatment is not ideal tissue regeneration.In the periodontal tissue regeneration process,gingival epithelial hyperplasia of the fastest,rapidly formed epithelial attachment,in the affected bone tissue regeneration and repair.Nymans proposed the guided tissue regeneration(Guided Tissue Regeneration, GTR)concept in 1982,and the basic principle is to place a surgical approach to selective physical barriers separating different periodontal tissue,that is to stop the source of gingival connective tissue fibroblasts.First of all cells and root surface contact with the membrane in a wedge-shaped space between the root and guide the periodontal ligament cells,the highest migration to the growth and formation of periodontal tissue regeneration.The GTR law to the treatment of periodontal disease in a new way,but simply GTR membrane only played a mechanical barrier and isolation role,to the hard and soft tissues GTR membrane is also active differentiation and accelerate growth,it is essential GTR to the joint use of growth factor.Recombinant human bone morphogenetic protein-2(recombnant human bone morphogenetic protein-2,rhBMP-2)is a relatively large number of studies in recent years,has a strong ability of bone growth factor-induced one.RhBMP-2 is the transforming growth factor-βsuperfamily member of the target cell growth,differentiation and apoptosis regulation and control role,in the periodontal tissue can be induced by undifferentiated mesenchymal cells to differentiate into cells into osteoblasts cementum Cell.The experimental research rhBMP-2 in the periodontal tissue regeneration in the lead role.Objective:To study the rhBMP-2 on the guide periodontal tissue regeneration, and to explore in the periodontal tissue regeneration in the lead role in the future and the prospect of clinical application.Methods:18 healthy male Wister rats were randomly divided into 3 groups,in the cheek before the gingival sulcus crown,1 mm for the experimental periodontal bone defects,conducted by Leo root surfaces were treated,the control group directly gingival flap reset,tight suture incisions,collagen membrane(GTR)in the defect placed collagen membrane,gingival flap reset, tight suture incisions.RhBMP-2,combined with collagen membrane(rhBMP-2) implantation defects in the rhBMP-2 after placing collagen membrane,gingival flap reset,tight suture incisions.4,6,8 respectively in the first week of the group killed two rats,10%formaldehyde cardiac perfusion,remove the mandibular(including the mandibular central incisors,gingival and alveolar bone),a 10%formalin-fixed,decalcified acid,ethanol dehydration gradient, embedded in paraffin,to the far produced nearly 5μm thick tissue,HE staining and Gomori special staining was observed under light microscope after periodontal tissue regeneration.Results:The gaps in the control group periodontal tissue regeneration of the worst,less periodontal tissue regeneration,a large number of epithelial defect of all.Application of the GTR of the two groups and the control group periodontal tissue regeneration significant difference,no gingival epithelial all long for bone regeneration reserved space;rhBMP-2-intensive group and trabecular bone osteoblasts,which showed that rhBMP -2 GTR joint operation to facilitate the repair of bone tissue that rhBMP-2 combined with GTR and the important role:①guide to the long bone tissue,biofilm can be improved simply insufficient to increase bone mass;②rhBMP-2 have a highly-efficient induced osteoblast activity and bone induction into the selection of new bone.The experiment of rhBMP-2 and GTR is the subject of the role of the experimental rats periodontal bone defects,and periodontitis from a variety of factors arising from the role of periodontal bone defects is not the same,and therefore the results might have some clinical effects of deviation in the rhBMP-2 GTR periodontitis in clinical repairing bone defects caused by the effect pending further study.
