Tumor seriously threat to human health and life, and the rateof tumor incidence and mortality increased year by year. It isimportant issue in the worldly medical profession that explores themechanism of tumor and develops the effective drug of treatmentof tumor. In the tumor treatment, except traditional surgery,radiotherapy and chemotherapy, it is the hot point to employtraditional Chinese medicine to integrated control treatment inrecent years. More and more foundations and clinical researchmaterials indicate that there are broad prospects antitumorly intraditional Chinese medicine, and mechanism antitumor totraditional Chinese medicine study from multi disciplinary tolaunch thoroughly multi level too.In traditional chinese medicine, tumorigenesis is the result ofdeficiency of blood and viscera dysfunction. The four basic ruleson the therapy of tumor by traditional chinese medicine ispromoting blood circulation by removing blood stasis , clearingheat and expelling miasma, softening hard lumps and dispel nodes and supporting healthy energy. It is extremely effective on thetherapy of tumor by the efficacy of promoting blood circulation byremoving blood stasis and resolving dampness by tonifying spleenof fructus polygoni orientalis.The microenvironment of tumor cell is composed of stromacells and extracellular matrix. Matrix metalloproteinase (MMPs) isone member in zinc peptidase, it is an indispensable enzyme incomposing connective tissue extracellular matrix degradationprocess, and it can almost degradation all the compositions ofextracellular matrix. MMPs has significant function in process oftumor metastasis. The expression and activation of MMPs iscorrelated with the growth,invasion and diversion of tumordirectly. MMPs is often expressed excessively in the malignanttumor. The great studies indicated that MMPs act on biologicalbehavior of tumor by the following three aspects:(1) Tumor cells are induced apoptosis or tolerance by MMPs.It is an important link in tumorigenesis that the tumor cells canescape apoptosis.(2)MMPs can promote angiogenesis of tumor. The tumor cells can propagate quickly depended on nourishment and oxygenprovided by lots of blood vessel organization. In the process ofblood vessel formation and tumor metastasis, both vascularendothelial cell and tumor cell can secrete MMPs to depredateECM. MMPs is important in the process tumor metastasis.(3)The chiefly barrier that arrest tumor metastasis is ECM.The present MMPs almost can depredate all ECM.(4)Adhering of substance and tumor cell is controlled byMMPs. If the process of tumor metastasis would successfullycomplete, the tumor cell must have the ability of adhering andavoid adhering. MMPs can influence the adhering of cell.A mountain of task of developing miracle drug used to treattumor awaits the medical community. In all these cases, MMPs isimportant in the tumor growth or development. Developing thenew drugs as the curative target of MMP/TIMP is becoming of thehot spot in interior and exterior exploitation. At present, severalkinds of broad spectrum and low molecular number of MMPscatastaltica are used in some kinds of tumor therapy are assessed inclinic. Although these macromolecule medicines can high performance inhibit activity of MMPs and prevent tumors'growthand diversion in preclinical animal experiments, their expensiveprice and fervent side effect in clinic lead to the failure in anaphaseclinic. Traditional Chinese drugs are purported to haveantineoplastic properties, and the research on the therapy of tumorby a sort of traditional chinese drugs has been going on in depth.Modern medical search results have proved that traditional chinesedrugs have the function of anticancer and mutation avoided,extremely effective and minimal side effects and low price.Voluminous data indicate that the fructus polygoni orientalis haveextremely effective to treat tumor. In the dissertation, themechanism of action of fructus polygoni orientalis inhibitingMMPs on the therapy of tumor is studied.Experimental purpose: The object of this experiment ispracticed in order to study whether fructus polygoni orientalis hasthe ability of res training MMPs. Then compared with the blankunit and positive control, theinhibition strength can be obtained.And the treatment of tumor by the fructus polygoni orientalis isindirectly responsed. Experimental principle and method: According to enzymedegradation of fluorescent substrate issued fluorescence principle,The inhibitory activity of fructus polygoni orientalis to MMPs bydrug design targeted on MMPs is studied in this paper. Thedetermination of inhibiting action is adding the fructus polygoniorientalis extractive in the reaction system. Thus, the inhibitingcompositions in the extractive cohere with MMP 16 and makingthe substrate shouldn't cohere with MMP 16, so the velocity ofdegrading substance lower. Then the status that the extractives offructus polygoni orientalis inhibit MMP 16 by the change offluorescence intensity is chalked up. The relation of concentrationof the fructus polygoni orientalis extractive and the velocity ofdegrading substance is observed by different concentration of thefructus polygoni orientalis extractive in different group. Theexperiments have two groups. One is purification fructus polygoniorientalis by chromatography and allocation liquor of differentconcentration. The other is detection changes of fructus polygoniorientalis extractive by different concentration inhibit MMP by themethod of fluorescent substrate. Experimental result: The experimental result proves, fructuspolygoni orientalis extractive have inhibitory on MMP 16activity,and the required amount of fructus polygoni orientalis atMMP 16 inhibit the activity of 50% is 0.2mg/ml, namelyIC50=0.2mg/ml. The concentration of the fructus polygoniorientalis extractive varies directly the velocity of degradingsubstance. Based on the foundation above, it is concluded thatfructus polygoni orientalis should inhibit MMPs, which probably isan important mechanism of action that the therapy of tumor byfructus polygoni orientalis.The traditional chinese drugs have the advantages that manytarget, many link and many effect, and its can affect many aspectsin tumorigenesis. It has attracted increasing attention. To interpretmechanism of cure for the disease by the traditional chinese drugs,the modern scientific method of biochemical, molecular biologyand immunology should applied in traditional Chinese drugs. Thenrational drug is designed to made experiments on animals andclinical trials. It make the application of traditional Chinese drugsso scientific, quantized, safe and rational that benefit the whole of mankind and make the traditional Chinese medicine integrate withthe world.
