Objective:To discover the possible effects of SDC-2 in the genesis of keloids. Correct theories then could be provided to prevent and treat keloids with SDC-2 and its correlation cytokines.Methods:All the samples were obtained from patients who visited the Department of Plastic Surgery,NO1,Affiliated Hospital of Anhui Medical University from 2006 to 2007.Thirty cases were used in IHC,includsing 15 keloids and 15 normal skins.All the patients were not suffering disease of adrenal gland,infectious diseases, dermatogic disease,immunological disease,infection and ulcer in local.They had not been under hormone therapy,immunodepressant therapy and local radiotherapy.Fresh samples were put into 10%neutro-formalin solution immediately for 24hrs then it came the dehydration and paraffin imbedding.Each sample was cut to 5μm sheets. Elivision method was used to dyeing the sheets.PBS was used as a substitute for the primary antibody on the negative control.The expression intensity and distribution of SDC-2 in the above 2 kinds of different tissues of 30 cases under 400 times light microscope were observed.Six high power fields were taken in each sheet.Statistics was used to analyze the data.Twelve cases were used in real-time PCR including 6 keloids and 6 normal skins.Cell culture was enforced by the method of literature. Transfer of culture was operated when cell fused.Fibroblasts of each sample were cultured during 5-6 generations of cells grew.Fibroblasts were dissociated after 48hrs. Their RNAs were extracted and cDNAs were reverse transcribed.The expression of SDC-2 mRNA was detected by using real-time PCR.Experiment on each sample was repeated for 3 times.Statistics was used to analyze the data of amplification to identify whether there was mRNA expression difference.Results:1.IHC Comparative study on expression of SDC-2 in normal skins and keloidsObserved under 400 times light microscope,SDC-2 expressed in epidermis and dermis of above two tissues.Yellow or buffy stains located mainly in the cells of epidermis.They could also be seen in the follicular epithelium cells and vascular endothelial cells.They expressed strongly in most keloids.The positive rate was 40.00%in normal skins,86.67%in keloids.The differences were significant between keloids and normal skins.2.Study on transcriptional levels of SDC-2 mRNA in Normal Skins and KeloidsStatistics analytic results showed that expression of SDC-2 gene in keloid fibroblasts was higher than in normal skin fibroblasts.The differences were obvious.Conclusion:SDC-2 play key roles in the signal transduction in the form of keloid. The strong experession of SDC-2 is close correlation with form of keloid and some tumors.Researches of SDC-2 and SDC family have important significance in preventing and treating keloid and tumors..
