Objective: Seminoma is a larvaceous and dangerous testicle malignant tumor. It originates from the germ cell. It is the most frequent malignant tumor. When the people have this disease,the death rate will become vary high if they don't have the treatment in time. So early discovery and treatment are the key of healing well the seminoma. As the other malignant tumors, the formation and the development of the seminoma are the result of multiple steps and multiple genes. tumorigenesis considers that an important component element of the tumorigenesis procedure is that all the kind of gene expressions have changed, the same meaning is that the proto-oncogene in cells have been activated and overexpressed, and the anti-oncogene has been deactivated. So, we investigated the oncogene and the anti-oncogene's change of the canceration procedure of seminoma from the molecular level by the genes' research of Oct4,PLAP,VEGF,p53, and so that we could provid the molecular biology foundation for the early identified diagnostic and treatment of seminoma.Methods: 38 paraffin specimen of seminoma were collected. IHC was used to detect the protein of Oct4,PLAP,VEGF,p53, saturated hydroxybenzene chloroform was applied to gain the DNA from 11 paraffin tissue and PCR-SSCP was used to examine the delation and mutation of p53 in seminoma.Results: (1)The positive expression rate of Oct4 in seminoma were 100%, in normal testicle organism were 0. the positive expression rate of PLAP in seminoma were 89.47%, in normal testicle organism were 0 too. However, on the base of the observation of microscope, there was little or no background staining in the all slices of Oct4, and there was a high level of background staining artifact in many slices of PLAP.(2)The positive expression rate of VEGF in seminoma clinical stageâ… ,â…¡,â…¢were 33.33%,90.91%,100%. There were different significance of VEGF between Stageâ… and stageâ…¡,stageâ… and stageâ…¢( p<0.01), there were no different significance of p53 between Stageâ…¡and stageâ…¢( p>0.05).10 normal testicle organism samples, 2 samples were weakly positive, 8 samples were negative.The positive expression rate of p53 in seminoma clinical stageâ… ,â…¡,â…¢were93.33%,54.55%,50%. There were different significance of p53 between Stageâ… and stageâ…¡,stageâ… and stageâ…¢( p<0.05), there were no different significance of p53 between Stageâ…¡and stageâ…¢( p>0.05).10 normal testicle organism samples were all negative.(3)There were no correlations among VEGF and p53 in seminoma (r =0.183, p>0.05).(4)There were p53 mutations in seminoma, the mutation rate was 45.45%.Conclusions: (1) Oct4 were all positive in seminoma, it is an excellent marker gene of seminoma diagnostic. It is more sensitive than PLAP. (2) with the development of seminoma clinical stage, the positive expression rate of VEGF were higher and higher, it suggested that VEGF played a very important role in genesis and clinical development of seminoma.p53 played the role in the earlier period of clinical stage in seminoma, there were no correlations among VEGF and p53 in seminoma. (3) p53 had the genes abnormality with the genesis and development of seminoma.
