Objectives: To study the effects of pPolyHb and Hb on the immune function of immunosuppressed mice induced by CTX and the dose-effect correlation.The immunosuppressive mouse model was established by using the chemotherapy drug CTX,which can simulate the side effects of immunological dysfunction caused by clinical CTX treatment of cancer.This study systematically studied the effects of pPolyHb and Hb on immune function of immunosuppressed mice induced by CTX in non-specific immunity,humoral immunity,cellular immunity,andSpleen histopathology.Methods: Establishment of a mouse model of immunosuppression: CTX(Cyclophosphate,CTX,100 mg/kg)was injected into the mice via the abdominal cavity for 2,5,and 6 days,respectively.According to the state of the mice and the basic indicators of immunity,5 days were selected as the optimal modeling time.1.pPolyHb and Hb were administered to the tail vein of mice at 7 mg/kg,1.4 mg/kg,and 0.7 mg/kg,respectively.The status,body weight,spleen index,and total number of peripheral blood leukocytes of mice were observed.The carbon clearance method was used to detect the phagocytic ability of mouse monocyte-macrophages,and to explore the effects of pPolyHb and Hb on non-specific immune function in immunosuppressed mice;2.pPolyHb and Hb were administered to the tail vein of mice at 7 mg/kg,1.4 mg/kg,and 0.7 mg/kg,respectively.The antibody-producing cell assay was used to determine the effect of pPolyHb and Hb on the humoral immune function in immunosuppressed mice;3.pPolyHb and Hb were administered to the tail vein of mice at 7 mg/kg,1.4 mg/kg,and 0.7 mg/kg,respectively.The levels of immune-related factors IL-1?,IL-2 and TNF-? in the serum of mice were detected.The mouse delayed-type hypersensitivity assay was used to determine the effects of pPolyHb and Hb on the cellular immune function in immunosuppressed mice;4.The pathological effects of Hb and pPolyHb on the spleen of immunosuppressed mice were investigated by observing the HE staining sections of spleen tissues of immunosuppressed mice.Results: This experiment showed that each dose of Hb can effectively enhance the immune function of immunosuppressed mice,while the effect of pPolyHb is not obvious: 1.Hb(1.4 mg/kg)can increase the number of leukocytes,the body weight,the spleen index,and the carbon clearance ability of immunosuppressed mice,which indicated the enhancement of non-specific immune function in mice.However,the effect of pPolyHb on non-specific immune function in mice was not obvious;2.Hb(0.7 mg/kg and 1.4 mg/kg)can increase the number of antibody-producing cells in immunosuppressed mice,which indicated the enhancement of humoral immune function in mice.The effect of pPolyHb on humoral immune function in mice was not obvious;3.Hb(1.4 mg/kg)can significantly increase the serum IL-1? level in immunosuppressed mice;Hb(1.4 mg/kg)and pPolyHb(1.4 mg/kg)can increase the serum TNF-? level in immunosuppressed mice;Hb(1.4 mg/kg)can also significantly increase the swelling rate of right ear in immunosuppressed mice,which indicated the enhancement of cellular immune level of mice.The experiment also found that pPolyHb and Hb did not significantly increase the serum IL-2 level in immunosuppressed mice.4.By observing the spleen tissue sections of immunosuppressed mice,it was found that the pathological changes in the spleen tissues of the CTX+NaCl group were obvious,and the immunosuppression of this group was serious;while the spleen lymphocytes in the CTX+Hb group were relatively activated.In the state,the number of lymphocytes increased,which promoted the immune function of immunosuppressed mice.However,the number of lymphocytes in the spleen of CTX+pPolyHb mice was not significantly increased,and its immune promotion effect on immunosuppressed mice was not obvious.
