Evaluation On The Immunomodulation Of Jiaweisangen Tang

Objective:Jiaweisangen Tang —a liquid pharmaceutical preparations,mainly prepared by American ginseng,fresh Baimaogen,fresh Lugen and Dandelion.Using the pharmacology research method from Chinese Materia Medical,the immunoregulation of Jiaweisangen Tang were explored,to provide a foundation of research for developing health food with the immune-enhancing function.Method:The health male mice were randomly divided into four groups(12 mice per group)— solvent control,low dose group,medium dose group and high dose.Low,medium and high doses were 5,10 and 30 times of the recommended amount of human,which is 1.3mL/kg,2.6mL/kg,and 7.8mL/kg,respectively.Jiaweisangen Tang were diluted into 200 mL stock solution by distilled water.Meanwhile,solvent control were treated with distilled water.20 mL/kg Jiaweisangen Tang was fed by gastric perfusion once a day,and administrated for 30 consecutive days.1.The humoral immune response of Jiaweisangen Tang was studied by body weight determination,cell quantity with antibody production,and serum hemolysindetermination,respectively.2.The cellular immune response of Jiaweisangen Tang was analyzed by delayed-type hypersensitivity experiment and adopting ConA induced mouse spleen lymphocyte transformation test,respectively.3.Innate immunity of Jiaweisangen Tang was surveyed by mice carbon clearance test and abdominal macrophage phagocytosis test,respectively.4.The NK cytoactive responsed to Jiaweisangen Tang was studied by NK cell activity detection.Results:1.Compared with control group,there was no difference in the body weight at the middle and the end of the experiment(P > 0.05),when dose changed from low to high.The quantity of plaque forming cells and HC50 were increased as dose increasing,and quantity of plaque forming cells in experimental group was much more than control group(P < 0.05).2.The swelling degree was increasing as the dose of Jiaweisangen Tang adding,and the swelling degree of experimental group was higher than control group,with statistical significance(P < 0.05).The transformation of lymphocyte induced by ConA was also increased as the dose of Jiaweisangen Tang raising.The date at high dosage group was significant higher than that in control group,with statistical significance(P <0.05).3.The capability of mice carbon clearance increased with dosage of Jiaweisangen Tang,compared with control group,high dose group has a remarkable effective,(P < 0.05).However,there was no difference on abdominal macrophage phagocytosis tests between experimental group and control group,P > 0.05.4.The activity of NK cell enhanced by Jiaweisangen Tang has a dosage-dependent effect,The ability of NK cell activities in the medium dose group and high dose group were all significantly higher than that in the control group,P < 0.05.Conclusion:1.Jiaweisangen Tang didn't affect the mice growth and body weight.However,it could significantly increase the quantity of plaque forming cells of spleen and HC50,and also promote the ability of producing antibodies and serum hemolysin at each experimental group,which indicated that Jiaweisangen Tang has the potential in enhancing humoral immune responses.2.All doses of Jiaweisangen Tang could remarkably increase the pedal swelling and the abilities of lymphocytes proliferation,and enhance delayed-type hypersensitivity and induce spleen lymphocyte transformation by ConA and further promote the ability of T cell and lymphocyte transformation.Jiaweisangen Tang has a certain ability to promote cellular immunity.3.There is no different in Spleen index and thymus index after mice were fed with Jiaweisangen Tang.High dosage group could enhance the capability of mice carbon clearance,and promote the ability of non-specific engulfment of reticuloendothelial system.The function of engulfing chicken erythrocytes by phagocytes may not be enhanced by Jiaweisangen Tang and nor monocyte-macrophage.4.NK cell activities induced by Jiaweisangen Tangare highly dependent on dosage,and also Jiaweisangen Tang showed a certain capability to promote non-specific immunity.In summary,Jiaweisangen Tang have the abilities in enhancing the immuno-function,and have the potential to develop as health products.
