Radix Scrophulariae is the dried root of Scrophularia ningpoensis Hemsl.,with the function of cooling blood and purging fire for removing toxin,which is used for consumption of YIN caused by febrile disease,dark red tongue and polydipsia,bodyfluid injury and constipation,over-strained cough,conjunctival congestion,pharyngalgia,crewels and diphtheria in the clinical.Modern medicine studies indicated:Radix Scrophulariae has various pharmacological action,such as antibacterial,anti-inflammatory and so on.For the past few years,it is used for curing chronic pharyngitis in the form of the compound recipe,which has the better effect.In this paper,the quality criteria of Radix Scrophulariae have been studied and the method for evaluating the quality of figwort has been established.Fist,some pharmacognosy identification of figwort have been carried out,such as the source,medicinal herbs traits,microscopic characteristics and TLC,then,the standard of pharmacognosy identification of figwort has been established.Which can provide scientific basis for the identification and development of this medicinal.Second,the comparison of cinnamic acid content has been measured among the samples from 14 different places with HPLC.As results,the range of content of cinnamic acid is from 0.0060%to 0.0783%,and the content of the sample from Hubei is the highest.So,cinnamic acid content should not be less than 0.02%.In addition,different processing methods of figwort samples form 5 different places in Wulong County,Chongqing Municipality were studied.The results showed that:fresh root,after steaming 48 hours,dried in the sun,effective constituent cinnamic acid is the highest.Which processed product is better pending combination of pharmacological and clinical research further identified.In addition,according to Chinese Pharmacopoeia(2005),the content of the quality evaluation of the system were detected,including water,the total ash and acid-insoluble ash,water-solubility and alcohol-soluble extract.All of content consistent with the Chinese Pharmacopoeia Radix Scrophulariae under the relevant regulations,which provide a basis for experimental and theoretical foundation for the establishment of quality standards Radix Scrophulariae,which also provided a guarantee for formulation of medical material quality standard. A method of HPLC-FP of Radix Scrophulariae was established.By studying comparatively the fingerprints of famous-region drug from Chongqing,determination condition of FP have been ascertained and 14 peaks have been identified as the general peaks.Among these peaks,the peak of cinnamic acid is taken as the standard one,and the relative retention time and relative peak area of the others are defined as parameters evaluating the quality.Which meet the specification of fingerprint research of TCM injection(Provisional)and the skill guide of empirical study of fingerprint research of TCM injection.At the same time,modeling object of famous-region drug(Radix Scrophulariae)from Chongqing have been screened and standard fingerprint model also have been synthesized by evaluating many active component of Radix Scrophulariae from Wulong County,Chongqing Municipality.The relative retention time and relative peak area of every characteristic principle were calculated by cinnamic acid as reference peak.In this way,Radix Scrophulariae from different place in Zhejiang were distinguished,it can track drug source from different place,ensure quality control of Chinese patent medicine and provide a foundation for establishment of quality control method.The method can be used to identify and evaluate the quality of Radix Scrophulariae.It regulated quality standards of Radix Scrophulariae,strengthened the quality control of Radix Scrophulariae and provided experimental evidence for the enactment of quality standards of Radix Scrophulariae.
