1. Xuanshen is the root of Scrophularia ningpoensis Hemsl., family Scrophulariaceae. It is an important traditional Chinese medicine which can nourish yin. The water-soluble components of Xuanshen mainly are iridoids and phenylpropanoid glycosides. The fresh roots of Xuanshen are white, but it will change to black after the traditional processing. Reported researches are all about the processing drugs, and the fresh roots of xuanshen are not much concerned. Here, we study the extraction and separation of the constituents in the fresh roots of xuanshen. By using column chromatography on macroporous resin, sillica gel and preparative HPLC, we got six chemical compounds, and the structures of them were authenticated by means of spectroscopy(UV,IR, 13C NMR,1H NMR,HMBC,HMQC, etc.).Four of them were iridoids. The results of extraction experiments show that the yield of aucubin is much higher than which was reported. It should be related to the different material which were used.In order to get the further message of the iridoids in xuanshen We also determined four of them ( aucubin, harpagide, 6-O-methyl-catalpol, and harpagoside) we have got in different samples of Radix Scrophulariae by HPLC using double wavelength and gradient elution. Through this research we have done some fundamental research about the changing and distribution of components in Radix Scrophulariae. The results show the content of all components in cut crude drug significantly lower than in the fresh root which were dried in the oven. And the content of harpagoside which was required assaying in the china PH is much lower than the content of harpagide. Besides, the content of aucubin in samples which were dried in the oven were much higher then those in cut crude drug. Meanwhile, By studying the migration behavior of the above four iridoids on capillary zone electrophoresis (CZE) and micellar electrokinetic capillary electrophoresis (MECC). We have done some preliminary research for using CE to determine the above four components which is much quickly. 2. Niubangzi is the dry ripe fruit of Arctium lappa L., family compositae. The maincomponent of it is arctiin whose content is about 4%—5%, and it also contain low quantitative arctigenin which shows strong pharmaco-activity such as anti-virus, anti-tumor and immunological regulation. This paper, we apply biological hydrolysis method to prepare arctigenin by enzymolysis of arctiin with synaptase. The results show hydrolysis completely according to TLC. Study on getting arctigenin from arctiin is important for the resource exploitation of Niubangzi.
