Relationship Of Interleukin-1β And Interleukin-1 Receptor Antagonist Gene Polymorphisms With Asthma

Objective: 1. To study the distribution of interleukin-1 gene polymorphisms in popu- lation in Jiangxi Province. 2. To study the relationship between IL-1 gene polymorph- ism and asthma development. 3. To study the relationship between IL-1 gene polym- orphism and concentration of IgE in asthma.Method : Genomic DNA was extracted from the peripheral blood of 105 asthma and 120 normal controls and then IL-1βand IL-1Ra gene were subjected to PCR amplification. The PCR product of IL-1βwas digested by restriction endonuclease AvaI. Both of the products were subjected to 2.5% gel electrophoresis and ethidiumbromide staining. The serum total IgE was examined with ELISA method. All statistics were analyzed by software SPSS 12.0.Result: In controls , the frequency of CC,CT and TT of IL-1β-511 were 34.2%,50% and 15.8%. AlleleⅠ,Ⅱ,ⅢandⅣof IL-1RN were found, and genotypeⅠ/Ⅰ,Ⅰ/Ⅲ,Ⅰ/Ⅱ,Ⅱ/ⅡandⅡ/Ⅳoccupied 74.3%,1% ,22.7%,1% and 2%. In asthma , the frequency of CC, CT and TT of IL-1β-511 were 21%,47.4% and 31.6%. AlleleⅠ,Ⅱ,ⅢandⅣof IL-1RN were found and genotypeⅠ/Ⅰ,Ⅰ/Ⅲ,Ⅰ/ⅡandⅡ/Ⅳoccupied 83.8%,1%,14.2% and 1% respectively. The frequency of IL-1β-511 TT genotype and T allele in asthmas was remarkably higher than in controls. The value of OR were 2.462,1.793 respectively. The frequencies of genotype and allele of IL-1RN were not statistically significant between the patients and controls. And there were no statistical difference among genotypes of IL-1(IL-1βand IL-1RN) and serum IgE level in asthma.Conclusion :1.The polymorphisms of the promoter region - 511C/ T of IL-1βgene are associated with asthma, T allele may be a risk factor for asthma. But this polymorphisms are not associated with the level of serum IgE. 2. The polymorphisms of the IL-1RN gene are not associated with asthma and the level of serum IgE.
