ObjectiveThis topic plans to use the technology of immunohistochemistry SP to detect the expression of SleX and E-Selectin in colonic carcinoma and lymphoid knot tissues and to probe whether Sialyl Lewis-X and E-Selectin protein correlate with patient's prognosis .The correlation of SleX and E-Selectin expression with the metastatic potential of human colonic carcinoma tissues was discussed.MethodAll the dates were analyzed by SPSS 11.0 statistical package, the count information calculated the positive rate, The comparison of positive rates uses the signed-ranked test. The relation of two variable are analyzed by the correlation analysis. The level of significant difference isα=0.05.Result1) Expression of the sialyl Lewis-X : (1) In the colonic cancer tissue the SleX expression was located in cytoplasm and cell membrane of the tumor. Normal mucosa tissue have no expression .(2) In 35 cases tissues, tumor with lymph node metastasis had more SleX protein expression(89.5%) than those without metastasis (43.8%) , P<0.01. (3)SleX expression in poorly differentiated group (85.7%) is higher than those in well-differentiated group(42.9%) , P<0.05;(4) Compared with without invasion serosal stratum group(55.0%), the SleX protein expression in serosal stratum invasion group(86.7%) increase significantly ,P<0.05. (5)According to liver metastasis, SleX expression in liver metastasis group(100%) had no difference with those without liver metastasis(85.7%),P>0.05. (6)Comprising the prognosis between the survival group in 3 years(52.9%) and death group in 3 years(93.3%) , the difference is also significant , P<0.01.(7)SleX expression in metastatic lymph node (94.7%) had no difference with those in primary position (68.6%), P>0.05,and it's higher than those in nomal lymph node (18.7%) , P<0.01 . SleX expression in primary position(68.6%) was higher than those in normal node (18.7%), P<0.01 . (8)Besides, the SleX expression had no relation to the patient'age, sex and differentiation,P>0.05.2) Expression of E-Selectin :(1)The expression of E-Selectin in the colonic cancer tissue mainly locate in blood vessel endothelium and lymph endothelium of tumor mesenchymal cell , the tumor cellular membrane and normal mucosa do not express. (2) In 35 cases tissues, tumor with lymph node metastasis had more E-Selectin protein expression(89.5%) than those without metastasis(25.0%),P<0.01. (3) E-Selectin expression in poorly differentiated group (81.0%)is higher than those in well-differentiated group(28.6%),P<0.05;(4)Compared with without invasion serosal stratum group(45.0%) , the E-Selectin protein expression in serosal stratum invasion group(80.0%) increase significantly , P<0.05. (5)According to liver metastasis , E-Selectin expression in liver metastasis group(100%) have no difference with those without liver metastasis(85.7%),P>0.05. (6)Comprising the prognosis between the survival group in 3 years (29.4%) and death group in 3 years(100%) ,the difference is also significant , P<0.01.(7) E-Selectin expression have no difference between in metastatic lymph node(68.4%) and those in primary position (60.0%) , P>0.05,and E-selectin expression in metastatic lymph node(68.4%) is higher than those in normal lymph node(12.5%),P<0.01; E-selectin expression in primary position (60.0%)was higher than those in normal lymph node (12.5%), P<0.05. (8)Besides, the E-Selectin expression had no relation to the patient'age, sex and differentiation,P>0.05.3)Correlation among the expression of SleX and E-Selectin in colonic carcinoma: There was not a significant correlation between the expression of SleX and E-Selection ,P>0.05.Co-expression of SleX and E-Selection had a more significant relationship with depth of invasion, lymph node metastasis and prognosis.Conclusions:1. There were over-expression of SleX and E-Selectin protein in Colon carcinoma.2. The expression of SleX and E-Selectin protein was significantly correlated with depth of invasion ,lymph node metastasis, liver metastasis and prognosis in Colonic carcinoma, but there is no difference between liver metastasis group and lymph node metastasis group .3. The agreement between the expression of SleX and E-Selectin was significant, and the co-expression of the SleX and E-Selectin protein was significantly correlated with depth of invasion, metastasis in colonic carcinoma.4. To detect the expression of SleX and E-Selectin protein will facilitate the colonic neoplasm severity estimation and its prognosis.
