Objectives:To investigate the role and mechanism ofα1,3 FucT-Ⅶon the metastasis potential of colorectal cancer.Methods:We investigated the expression ofα1,3 FucT-Ⅶin 30 clinical samples from colorectal carcinoma patients by RT-PCR and immunohistochemistry.Theα1,3 FucT-Ⅶ-high-expression colorectal cancer cell was established using human colorectal cancer cell line LOVO.Cell proliferation was measured by CCK-8 assay. Cell cycle was analyzed by flow cytometry.The alteration of LOVO cells metastatic potential before and afterα1,3 FucT-Ⅶgene transfection were assayed by cell chemotactic migration and invasion.Serial resection of colorectal cancer tumor was investigated with immune histochemistry of CD24 andα1,3 FucT-Ⅶ,and the expression change of glycoprotein CD24 and carbohydrate antigen SLe~X after stable transfection of LOVO was also examined in vitro.Results:Higher mRNA and protein expression ofα1,3 FucT-Ⅶwere observed in colorectal tumors compared with adjacent normal tissues.There was a positive correlation betweenα1,3 FucT-ⅦmRNA expression and lymph node status (p=0.018) and distant metastasis(p<0.05),andα1,3 FucT-Ⅶprotein expression was correlated to lymph node status(p=0.042) and Grade stage(p=0.022).The cell chemotactic migration and invasion were enhanced afterα1,3 FucT-Ⅶgene transfection in LOVO cells in vitro.Overexpression ofα1,3 FucT-Ⅶpromoted the expression of CD24 and SLe~X in LOVO cells.Conclusions:Our findings suggest thatα1,3 FucT-Ⅶwas correlated to lymph node meatstases in colorectal cancer.Andα1,3 FucT-Ⅶcould promote the expression of glycoprotein CD24.
