Daphne odora var. marginata, family of Thymelaeaceae, is a famous flower and medical plant in the regions along the Yangzi River. Resent pharmacological studies indicated that Daphne odora var. marginata had anti-lipid peroxidation, analgesic, immunoloregulation, anti-inflammation, antibacterial, anti-hypoxia and anti-tumor bioactivity. In the resently thesis, separation and purification the chemical constituents and antioxidant activity of Daphne odora var. marginata were studied. Hopes would offer the theoretical foundation for the depth development and utilization of Daphne odora var. marginata. Five chapters were carried on the elaboration in this thesis and the main contents were as follows:In first chapter, a review was presented, in which introduced the progress of chemical constituents, pharmacological activity and the technology of separation and purification the activity compounds of the plants in Daphne genus. The progress of chenmical constituents and pharmacological acticity of Daphne odora var. marginata were reviewed, too.In the second chapter, a counter-current high-speed extraction method was used to Daphne odora var. Marginata. Systematic solvents were employed to enrich chemical constituents. The extracts were analyzed by high-performance liquid chromatography (HPLC). Results approved that the counter-current high-speed and systematic solvents extraction method was useful in extracting the chemical constituents from Daphne odora var. marginata.In the third chapter, daphnetin was separated and determinated in various extracts from Daphne odora var. marginata by RP-HPLC. The main constituents varied between the extracts. The method was sensitive, reproducible and rapid.In the fourth chapter, an orthogonal L9 (3)4 test design was applied to optimize parameters including separation temperature, flow rate of the mobile phase and revolution speed of the apparatus. And the isolation parameters were obtained that: the lower phase of the two-phase system was used as the mobile phase at the flow rate of 1.8 ml/min, and the isolation temperature and revolution speed were set at 31℃and 850 rpm, respectively. Four high-purify compounds were successfully separated and purified from ether and ethyl acetate extract of Daphne odora var. marginata by HSCCC. The method was sensitive, selective, simple, rapid and low consumption. It will provide an advance technical for further analysis of chemical constituents of Daphne odora var. marginata.In the fifth chapter, the structures of the chemical constituents obtained from Daphne odora var. marginata by HSCCC were identified by UV, IR, ESI-MS, EI-MS, 1H NMR and 13C NMR. The result showed that they were daphnetin, daphnodorin I, p-hydroxybenzonic acid and daphnodorin D1.
