The genus daphne, which belongs to the Thymelaeaceae family and consists of around 44 species, was distributed in the southwest and northwest regions of China. Most of them were used as anti-inflammatory and analgesic agent in folk medicine. The stems and barks of some plants in daphne genus (Thymelaeaceae), commonly called"Zu Shima", is widely distributed in Shanxi, Gansu, Ningxia, Sichuan and Qinhai provinces of China. So far, several diterpenoids, flavonoids, biflavonoids, coumarins, bicoumarins, lignans have been isolated from this herb. It has been used in Chinese folk medicines to treat aches and rheumatism. In modern pharmacological investigation, it was used to treat toothache, waistache, rheumatoid arthritis, quadriplegia, thrombosis, inflammatory, and cancer et. al.In this dissertation, we investigate the chemical constituents from the stem bark of Daphne pedunculata H. F. Zhou ex C. Y. Chang systematically, anti-inflammatory and analgesic activity. Forty-five compounds were isolated from the stem bark of Daphne pedunculata by silica gel, Sephadex LH-20 and polyacylamine chromatography and thirty-four compounds were identified as follows by spectral analysis: CG-1:β-sitosterol CG-2:daucosterol CG-3:p-hydroxybenzoic acid CG-4:ethyl 4-hydroxybenzoate CG-5:n-dodecane CG-6:daphnetin CG-7:7-methoxy-8-hydroxycoumarin CG-8:1-dodecanol CG-9:7-methoxycoumarin CG-10:umbelliferone CG-11:octadecanoic acid CG-12:daphnoretin CG-13:2-tritetracontylene CG-14:n-octacosanol CG-15:daphneticin CG-16:isodaphneticin CG-17:(?)-pinoresinol CG-18:horsfieldin CG-19:syringaresinol CG-20:daphnenone CG-21:apigenin CG-22:daphneolone CG-23:luteolin CG-24:genkwanin CG-25:syringin *CG-26:3-[(3-hydroxy-4-ethyl propanpicatephenyl)oxy] -6-methoxy-7-hydroxycoumarin CG-27:daphnetin-8-O-β-D-glucoside CG-28:daphnorin CG-29:skimmin CG-30:daphnorin CG-31:luteolin-7, 3'-dimethylether-5-O-β-D-glucoside CG-32:yuanhuanin CG-33:glucogenkwanin CG-34:yuankaninAmong them, CG-26 was a new compound,and others were isolated from this plant for the first time.The crude extract, four extractive fractions and six compounds from this plant were evaluated for the anti-inflammatory and analgesic activities. the results of anti-inflammatory activity showed that the crude extract, EtOAc fraction, n-BuOH fraction, PE fraction, and compounds CG-6, CG-10, CG-22 displayed significant anti-inflammatory activity in vivo with asprin as control. And the crude extract, PE fraction, and EtOAc fraction, n-BuOH fraction showed analgesic activities.
