Seeding Of EPTFE With Endothelium Cells Induced From Rat Marrow Mesenchymal Stem Cells In Vitro

Objective:Application of rat bone marrow-derived mesenchymal stem cells from the vascular endothelial cells attached ePTFE wall,and joined the common culture with heparin for observation on the efficiency of cell-attached,and discussion the methods of improving the cells attaching.Methods:1.The marrow of thighbone in rats were collected with the density gradient centrifugation and cultured into the rarefied MSCs.Then differentiation into endothelial-like cells by cultivation in the presence of 10 ng/ml vascular endothelial growth factor(VEGF) and 2ng/ml basic fibroblast growth facotr(bFGF).2.The cells were characterized by immunohistochemistry stain,and observationâ…§factor associated antigen expression.And cells on the 14d were collected for observation with electronic microscopy of ultrastructural characteristics.3.Then 4 groups of the cells cultivated in ePTFE with the heparin sodium which concentrations were 0,10ug/ml,20ug/ml,30ug/ml.and detected the cells attaching rates every 4 hours.Results:1.Through the acquisition of density gradient centrifugation,sufficient MSCs were were ptoliferated in vitro,after immunohistochemical staining,the expression of vWF was positive in the induced cells.Cells of devited group demonstrated specific characteristics by transminssion electronic microscopy observation,espcially W-P bodies(Weibel-Palade bodies) were boserved.2.The rate of the cell attaching were increase gradually,and the 4 groups highest rate were 88.34%,96.48%,91.98%,85.69%at the piont of 20th hours.3.The heparin sodium could remarkable increase the rate of the cell attaching,and the best of concentration was 20ug/ml.Conclusion:The purified MSCs could be obtained and induced directionally into endothelium cells in vitro,and the heparin sodium could remarkable increase the effect of the cell attching.
