Objective:To detect human papillomavirus(HPV) 16,18 types in tissues and peripheral blood of cervical carcinomas and chronic cervicitis,and to explore the relationship between HPV infection and cervical carcinomas and the expression in tumour micrometastase.Methods:Real-time fluorescence quantitative PCR(FQ-PCR) was used for the detection of HPV 16 DNA and HPV 18 DNA in 48 cases of cervical carcinoma and 24 cases of tissues near to carcinoma and 40 chronic cervicitis and 20 cases of normal cervical epithelium tissue.Reverse transcription polymerase chain reaction(RT-PCR) were used for the detection of HPV mRNA in 96 case of peripheral blood of cervical carcinomas,and in 40 case of tissues and peripheral blood of chronic cervicitis.Result:Among 48 cervical carcinomas(Ⅰ-Ⅱb ),the positive rate of HPV-DNA 16 type is 66.7%.The average copy numbers of DNA template in 16 type positive samples was 3.27×10~5.The positive rate of HPV-DNA 18 type is 29.2%,the average copy numbers of DNA template in 16 type positive samples was 2.61×10~4.Among 96 cervical cance peripheral blood,the positive rate of HPV mRNA 16 type is 2.1%.HPV 16 DNA was positive in 14 cases of 24 cases tissue near to carcinoma(60%),The average copy numbers of DNA template was 1.85×10~4.Three out of 40 chronic cervicitis tissue HPV 16 DNA was positive(15.0%),Each of tissues and peripheral bloods HPV 18 was negative.In 20 cases of normal cervical epithelium tissues and peripheral bloods,HPV DNA and mRNA was negative.The difference was markedly significant among the three types tissues.Conclusion:The infection of high risk types HPV is highly associated with cervical carcinomas.Real-time fluorescence quantitative polymerase chain reaction maybe an effective method in detection of HPV 16,18 infection and early diagnosis of cervical carcinoma.The detection of HPV mRNA 16 in peripheral blood of cervical carcinomas was low,and its clinical significance needs further evaluation.
