The Experimental Study On The Effect Of Drug Combination To The Kidney In Rabbit

Objective:To observe the affluence of drug combination on the kidney in rabbit and discuss the previous inspect value of ultrasound in acute renal failure(ARF).Method:Divided Zealand white rabbits into 4 groups,Gentamycin were injected intravenously at the dose of 160mg/kg to the Gentamycin group 1 hour,Gentamycin group 2 hours,Gentamycin group 3 hours.Levofloxacin were injected intravenously at the dose of 0.05mg/kg to the Levofloxacin group 1 hour, Levofloxacin group 2 hours,Levofloxacin group 3 hours.Drug combination were injected intravenously to the drug combination group 1 hour,drug combination group 2 hours,drug combination group 3 hours.Then take the blood to make biochemical measure,meanwhile the color ultrasonic examinateation was performed to measure the renal size and the hemodynamic changes and observe the blood perfusion of the kidney during ARF.Finally take the tissue to make specimen. Result:1,Ultrasound show:the renal size of Gentamycin group was significantly larged and the echo of renal cortex was enhanced,The blood perfusion of the kidney decreased gradually,compared with the control group,there was significant difference in length,width,thickness of the kidney and the resistive index,pulsatility index and the blood flow velocity of renal arteries of Gentamycin group P＜0.05.There was significant difference in the resistive index,pulsatility index and the blood flow velocity of renal arteries in drug combination group 3 hours between the control and the drug combination group 1 hours,2 hours, P＜0.05.2,Blood biochemical rheasure:the BUN and the Cr level of the Gentamycin group were increased significantly,compared with the control group, P＜0.05.The CK level of the drug combination increased significantly,compared with the control group,P＜0.05.There was significant difference in the BUN and Cr level in drug combination group 3 hours between the control and the drug combination group 1 hours,2 hours,P＜0.05.3,Light microscope:the renal structure changed significantly in the Gentamycin group,the renal glomerulers were shrink,the renal capsule expanded and the epithelia of proximal tubule were diffusely necrotic.The structure of the kidney was normal in the Levofloxacin group.The renal tubule changed mildly and the skeleton of psoas muscle broken and crossing band disappeared.Conclusion:(1) The acute renal failure(ARF) was induced by Gentamycin,whereas drug combination decreased the renal injury and induced a new side effect.(2)Ultrasound can be as a useful way to detect the renal size,the blood perfusion and the blood hemodynamic changes,further to inspect the ARF induced by drug.
