Objective:Research on environmental estrogens octylphenol and nonylphenol on the female reproductive system toxicity and oxidative damage to the liver in rats,and the intervention on octylphenol and nonylphenol with Pueraria lobata isoflavones(PLIs).To discuss the combined toxic effects of octylphenol and nonylphenol. Methods:130 adult female rats were randomly divided into 13 groups, each with 10. They were exposed to exposure group, the intervention group and the control group by gavage. Exposure group was divided into: 80 mg / kgOP, 320 mg / kg OP, 50 mg / kg NP, 200 mg / kg NP,80 mg/kgOP+50mg/kgNP,320mg/kgOP+200mg/kgNP.Intervention group was divided into: 200mg/kg NP+0.5% PLIs, 320 mg/kgOP+0.5% PLIs,320 mg/kgOP+200mg/kgNP+0.5% PLIs, 200mg/kg NP+5% PLIs, 320mg/kg OP+5% PLIs, 320mg/kgOP+200mg/kgNP+5% PLIs. After 60 days, they were copulated to healthy male rats and delivered naturally. Mather rats were put to death after ablactation. The reproductive action was observed, and stusdy the changes of hormone levels, oxidative damage in the liver, NOS activity in Uterine, as well as pathological changes in ovarian tissue. Results:Contact with the higher dose of octylphenol and nonylphenol, the weight and the coefficient of ovarian has no significant differenc(eP>0.05), the activities of NOS in Uterine has no significant difference(P>0.05), but exposure group exhibited a downward trend.Exposure group compared with the control group ,there were significant differences on FSH, LH, and E2 (P<0.05), shows a declining trend, there were significant differences on serum T(P<0.05), shows an upward trend. The high dose exposure group compared with the control group, there were significant differences on MDA content and the activity of SOD(P<0.05). Intervention group compared with the relative exposure group, there were significant differences on serum FSH, LH, T, E2 levels ,and MDA content(P<0.05).The combined exposure group compared with the relative low and high dose of octylphenol and nonylphenol, there were significant differences on serum FSH, T, and the content of MDA. Histopathological observation shows that the ovarian tissue apparent pathological changes. Conclusion:Octylphenol, nonylphenol and the combined group impact on the reproductive system of the female rats, the mechanism may be Serum FSH, LH, E2 decrease gonadotropin and the inhibition of steroid hormones, inhibit ovulation and cause reproductive dysfunction. MDA content increased, the activity of SOD declined, show that the two toxic caused oxidative damage on the liver. PLIs played a certain interventions on the toxic effects of these poison.
