Objective:Using modern research methods and techniques to study the quality of dispersible tablets containing isoflavones from pueraria lobata. Established qualitative and quantitative detection methods of the main components.By observing the preventive and therapeutic effect of dispersible tablets containing isoflavones from pueraria lobata on osteoporosis in rats ovariectomized-induced, identified the pharmacodynamic effects as well as mechanisms and molecular pathways played in the prevention and treatment of osteoporosis.Methods:Using TLC、HPLC and UV studied the quality of dispersible tablets containing isoflavones from pueraria lobata. Observed the preventive and therapeutic effect of dispersible tablets containing isoflavones from pueraria lobata on osteoporosis by ovariectomized rat model that was surgical removal of both ovaries, according to the weight randomization them after they were surgery three days. Intragastric administered dispersible tablets containing isoflavones from pueraria lobata200mg·kg-1,100mg·kg-1and50mg·kg-1every day,and set the model group, sham operation group. Six months after the administration, the rats were measured mechanical properties of femur by using external pressure method. Measured bone mineral density in vivo by dual-energy X-ray absorptiometry (DEXA).Determined serum estrogen levels and other indicators by radioimmunoassay and enzyme-linked immunosorbent assay. Results:The method for the determination of dispersible tablets containing isoflavones from pueraria lobata was highly accuracy, simple, easy to operate, stable and feasible. After six months of continuous administration, observing the indicators examined in each group of rats, suggesting that dispersible tablets containing isoflavones from pueraria lobata can significantly improved the general situation of the model rat. All three doses could significantly reduced the degree of body weight gain in the model rats and improved uterine factor, and three doses show good dose-effect relationship trends. Three treated groups were significantly improved resistance to external force of bone tissue, and the impact of the high dose groupwas the most obvious. Collected and calculated the ratio of wet weight, dry weight, ash weight and volume got from one side femur of the rats, suggesting that all the three groups could improved the performance of osteoporosis caused by modeling to some extent, and increase the content of bone calcium. Observing the serum indicators and found that the dispersible tablets containing isoflavones from pueraria lobata can increased serum estrogen levels. It can improve the contents of the serum bone-specific alkaline phosphatase (BAP),osteocalcin (BGP)and25-hydroxyvitamin D (25-HVD). It also can reduce the contents of serum tartrate-resistant acid phosphatase5b (TRAP5b) and type I collagen C-terminal peptide (CTX-I). The results reflected that this dispersible tablets can promoted the funtion of osteoblasts and inhibited the role of osteoclasts. It also can enhanced the content of bone calcium absorption and bone collagen, improved bone density, thus inhibited the further development of osteoporosis. Pathological observations further validate this mechanism of this molecular biology and medical effect of path.Conclusion:Dispersible tablets containing isoflavones from pueraria lobata may had estrogen-like activity, and played an important role in compensating for the absence of body estrogen. The dispersible tablets containing isoflavones from pueraria lobata had significantly increased serum levels of bone formatiom markers and reduced the serum levels of bone resorption markers. These results suggest that this preparation had the role of promoting osteoblast activity and inhibiting osteoclast activity, thus inhibiting the further development of osteoporosis. This study not only observed the efficacy of the characteristics on anti-osteoporosis of the dispersible tablets containing isoflavones from pueraria lobata, but also explored its biological mechanisms and phannacodynamic pathways through more than one index at different levels.
