Objective:To investigate the expressions of BRCA1 members of DNA repair gene family in patients with advanced non small-cell lung cancer (NSCLC), the response to platinum-based chemotherapy as well as their clinical prognostic significance. Methods: Expression levels of BRCA1 were detected by method of immunohistochemistry in 81 advanced NSCLC patients.Follow-up investige their treatment, therapeutic reaction, and progression time,survival time as well. Chi-square criterion, Kaplan-Meier survival curve were used for statistical analysis. Results: The masc rate of BRCA1 gene in advanced NSCLC patiences is 51.9%.There were no difference between different pathological classification and clinical TNM stages,age, saxuality, smoke state neither. Response rate of full set after platinum-based chemotherapy was 43.2%. control rate was 88.9%. While response rate of negetive BRCA1 is 56.4%, masc BRCA1 is 31.0%.The response rate of negetive BRCA1 higher than masic BRCA1. There was significance differrence between them through statistics analysis. The median progression time of disease of the patients with low expression of BRCA1 was 220 days.and the median- survival time was 538 days, one year survival rate was 54.8%. While the mid progression of disease of the patients with high expression of BRCA1 was 177 days.and the mid survival time was 352 days, one year survival rate was 30.8%..One year survival rate was make a significance difference between the two group.The expression of BRCA1 was make no differrence in survival time and the mid progression time of disease. Conclusion: The expression levels of BRCA1 make no difference with clinical feature. Patients with low expression of BRCA1 could make a successful chemotherapy. One year survival rate was higher, But no difference in total survival. Patients with low expression of BRCA1 maybe had better prognostic. BRCA1 could be used as predict to platinum-based chemotherapy response of NSCLC.
