Objective:Investigate the expressions of mismatch repair genes hMSH2, hMLH1andantioncogene P53in breast cancer tissues and normal breast tissues, and the relationshipamong these genes with clinicopathologic parameters, and study the correlation andclinical significance of hMSH2, hMLH1and P53.Methods:1. Select60paraffin pathology specimens of primary breast cancer tissues and normalbreast tissues during the period from2008.3to2014.8. The patients underwent breastcancer modified radical mastectomy surgery. No patients had received preperativechemotherapy or radiationtherapy, including55breast invasive ductal carcinoma,2breastinvasive lobular carcinoma,2breast mucoid adenocarcinoma,1breast invasivemicropillary carcinoma. The PTNM stage was I-III. To detect the expressions of hMSH2,hMLH1and P53protein in cancer tissues and normal breast tissues, and the expressions ofER, PR, Her-2in cancer tissues by immunohistochemical staining technology.2. Statistical analysis: Using SPSS19.0software for statistical analysis. UsingChi-square test or Fisher exact test, P <0.05was considered statistically significant,andusing Kappa test to inspect consistency between the expressions of hMSH2, hMLH1protein and P53protein.Result:1. In60cases of breast cancer tissues hMSH2, hMLH1, P53protein positiveexpression rate was68.33%(41/60),71.67%(43/60),48.33%(29/60), normal breast tissueshMSH2, hMLH1, P53protein positive expression rate was86.67%(52/60),90%(54/60),16.67%(10/60), hMSH2, hMLH1protein positive expression rate in cancer tissues waslower than that in normal breast tissues, P53protein positive expression rate in cancer tissues was higher than that in normal breast tissues, with statistical difference(P<0.05).2. In60cases of breast cancer tissues the positive expressions of hMSH2and P53protein had statistical difference with tumor size, axillary lymph node metastasis(P<0.05),while no statistical difference with age, PTNM stage, ER, PR and Her-2(P>0.05).3. In60cases of breast cancer tissues the positive expression of hMLH1protein hadno statistical difference with age, tumor size, axillary lymph node metastasis, PTNM stage,ER, PR and Her-2(P>0.05).4. There is moderate consistency between the expression of hMSH2protein andP53protein (Kappa=0.407,P<0.05). There is poor consistency between the expression ofhMLH1protein and P53protein (Kappa=0.183,P>0.05).Conclusion:1. hMSH2and P53have significant correlation with breast cancer’s occurrence anddevelopment, increase tumor’s malignacy degree.2. Detecting the expressions of hMSH2and P53can provide new clues for the stageand prognosis of breast cancer.3. hMLH1has significant correlation with breast cancer’s occurrence, no significantcorrelation with breast cancer’s development.
