Objective: To investigate the inhibiting effects of Celebrex alone and the combination with cisplatin , hydroxycampothecin on the proliferation and apoptosis-induced of the tongue squamous cell carcinoma line Tca8113,and to explore the anticancer mechanism of Celebrex.Methods: The effects of Celebrex and the synergistic effects of Celebrex in combination with cisplatin, hydroxycampothecin on proliferation of tongue squamous cell carcinoma line Tca8113 were detected by MTT assay. Apoptosis were detected by TEM (transmission electron microscope ),agarose gel electrophoresis and the Flow Cytometry. The datas were analyzed by SPSS13.0 general linear models procedure to determine the effects of each drug alone or combination.Results: With the telescope ,we observed the morphological changes of apoptotic Tca8113 cells in Celebrex groups.The Celebrex could inhibit the proliferation of Tca8113 cells by the manner of dose-dependent and sequence-dependent by MTT assay. The Celebrex could induce Tca8113 cells to apoposis.With the concentration of Celebrex increasing,the ratio of cells in G0/G1 phase increased and in S phase decreased. The lower concentration of Celebrex in combination with various concentration of cisplatin or hydroxycampothecin resulted in their reduction of IC50.The apoptotic rate in the combined groups was significantly higher than those of the control groups (p < 0.05). There was a positive correlation between inhibition ratio or apoptosis rate and concentration in the combined groups.Conclusion: Celebrex could inhibit the proliferation of tongue squmous cell carcinoma line Tca8113 and induce its apoptosis by the manner of dose-dependent and sequence-dependent.Celebrex could make Tca8113 cells be arrested in G0/G1 phase. Synergistic inhibiting effects were found when Celebrex in combination with hydroxycampothecin more than with cisplatin.
