Objective The purpose of this study was to investigate the expression of TFF1,COX-2,VEGF,KDR and micro-vessel density(MVD)in primary gastric carcinoma and adjacent carcinoma and to explore their role in carcinogenesis and metastasis of gastric carcinoma.Methods The expression of TFF1,COX-2,VEGF,KDR and MVD (CD34 monoclonal antibody labeling)were examined by S-P immunohistochemical method in 66 gastric carcinoma and 66 adjacent carcinoma.To use pathological image analysis system to analysis the grey level of TFF1,COX-2 and the positive cell rate of the VEGF,KDR and MVD values.Results The expression of TFF1 in gastric carcinoma is lower than that in adjacent carcinoma(P<0.01),The expression of COX-2,VEGF and KDR in gastric carcinoma is higher than that in control group(P<0.01).The MVD values in gastric carcinoma is higher than that in control group(P<0.01).There was close correlation between the expression of TFF1,COX-2 and differentiation and lymph node metastasis of gastric carcinoma(P<0.05).The positive cell rate of VEGF was higher in cases with poor differentiation,serosal invasion and lymph node metastasis than differentiated cases and cases without serosal invasion and lymph node metastasis(P<0.01).The expression of KDR have no correlation with age,sex,differential degree,serosal invasion and lymph node metastasis(P>0.05).There were also correlation between MVD and differential degree,serosal invasion(P<0.01).The expression of TFF1 in VEGF, KDR positive expression group was significantly lower than that in VEGF,KDR negative expression group(P<0.01).The expression of COX-2 in VEGF,KDR positive expression group was significantly higher than that in VEGF,KDR negative expression group(P<0.01).The MVD values in VEGF,KDR positive expression group was significantly higher than that in VEGF,KDR negative expression group(P<0.01).There was a negative correlation between the expression of TFF1 and COX-2(r=-0.891,P<0.01).The grey level values of TFF1 and MVD values was consistent(r=0.811,P<0.01).There was a negative correlation between the grey level values of COX-2 and MVD values(r=-0.737, P<0.01).Conclusions The expression of TFF1 in gastric carcinoma is lower than that in adjacent carcinoma.The result hints TFF1 may be a specific restrain factor to gastric carcinoma.The expression of COX-2,VEGF,KDR and MVD values in gastric carcinoma were consistent,it means COX-2,VEGF and KDR may have close correlation with tumor vessel neogenesis.COX-2 maybe mediates the interaction between TFF1 and vascular endothelial cell which induce neovascularization.
