Objective:To investigate the expression of Raf kinase inhibitor protein and nuclear factor-kappaB p65 and their relationship with clinical-pathologic parameters in breast carcinomas,and to approach the correlation between RKIP and nuclear factor-kappaB p65.Methods:The expressons of RKIP and NF-κBp65 were studied in 54cases of breast carcinomas tissue,22 cases of adjacent noncancerous breast tissues and 14 cases of hyperpiasia of mammary glands tissues by immumohistochemical technology(S-P). Results were compared with clinical-pathologic parameters,and the correlation between RKIP and NF-κBp65 was analyzed.Results:1.The immunostaining of RKIP localized mainly in cytoplasm and cytolemma.The positive expression rate of RKIP was significantly lower in breast carcinoma tissues than that in adjacent noncancerous breast tissues or hyperpiasia of mammary glands tissues(p<0.05).and the rates were 51.9%(28/54),81.8%(18/22)and 100%(14/14),respectively.The positive expression rate of RKIP was significantly lower in breast carcinomas with axillary lymph node metastasis than that in breast carcinoma without axillary lymph node metastasis(p<0.05),and it did not correlate with age,tumor size,histological grading,TNM staging,ER.et al(P>0.05).2.NF-κBp65 showed cytoplasm and/or nucleus staining.The positive expression rates of NF-κBp65 in breast carcinoma tissues,adjacent noncancerous breast tissues and hyperpiasia of mammary glands tissues were 42.4%(24/54),13.6%(3/22) and 7.14%(1/14),respectively.Significant difference was observed between the positive expression rate of NF-κBp65 in breast carcinoma tissues and that in adjacent noncancerous breast tissues and in hyperpiasia of mammary glands tissues(p<0.05). There was no significant difference between the expressions of NF-κBp65 in adjacent noncancerous breast tissues and that in hyperpiasia of mammary glands tissues(P>0.05).The expression of NF-κBp65 in breast carcinoma has a significant correlation with histological grading,TNM staging and axillary lymph node metastasis and ER(P<0.05).and it did not correlate with age,tumor size(P>0.05).3.There was inverse correlation between the expressions of RKIP and NF-κBp65 in breast carcinoma(P<0.05).Conclusions:1.There is down-regulation of RKIP in breast carcinoma tissues, The level of RKIP expression in breast cancer with lymph node involvement was significantly lower than in breast cancer without lymph node involvement. Down-regulation of RKIP may be implicated in the carcinogenesis and metastasis of breast carcinoma.2.There was negative correlation between the expressions of RKIP and NF-κBp65 in breast carcinoma.It is possible that RKIP locates the upstream of NF-κBp65 and plays an important role as a regulator of NF-κBp65.RKIP and NF-κBp65 may be implicated in the carcinogenesis of breast carcinoma together.
