ObjectiveNuclear factor-kappaB(NF-κB) is a key factor which can regulate genetic transcription of cells.It is retained in the cytoplasm by combining the inhibitor of NF-κB when resting.The activated NF-κB can translocate into nucleus and followed by modification,such as phosphorylation.Then it can combine with specified DNA sequence,have an effect on downstream factors,promote and regulate genic transcription which is associated with immune and inflammatory reaction,cell proliferation and so on.NF-κB p65 is a subunit of NF-κB.It has a great meaning on the transcriptional activation of NF-κB.Phospho-NF-κBp65 is a protein that has transcriptional activity.Ameloblastomas(ABs) are common epithelial odontogenic tumors,whose growth has a characteristic of locally invasion.The occurrence development and invasion of ameloblastomas are on the basis of the molecular biology.This study aims to approach the expressions of NF-κBp65 and Phospho-NF-κBp65 in human ABs,further,to know the correlation between the two proteins and the biological behaviour of human ABs and the role of both in the occurrence and development of this tomour.Materials and MethdodsA total of 103 ABs immunohistochemistry specimens(including 60 primary ABs,39 recurring ABs and 4 canceration ABs) were collected from Affiliated Stomatology Hospital and the First Clinical Hospital of China Medical University.96 ABs located in mandibula,5 ABs located in maxillary bone,and 2 ABs located in gum. The histological classifications of ABs include:87 solid types(29 follicular patterns,40 plexiform patterns,8 acanthomatous types,1 granular cell types,9 basal cell types),12 unicystic types,2 peripheral types,2 desmoplastic types.22 normal oral mucosas were collected.NF-κBp65,Phospho-NF-κBp65 and the inhibitors of NF-κB—IκBα,IκBβwere detected by streptavidin peroxidase method and avidin-biotin peroxidose complex technique of immunohistochemistry using monoclonal mouse anti-human NF-κBp65 and IκBαantibody,polyclonal rabbit anti-human Phospho-NF-κBp65,IκBβantibody.Results NF-κBp65 was expressed in the plasma of peripheral columnar or cuboidal cells and central polyhedral cells of odontogenic epithelium in ABs.The expressions of NF-κBp65 were most moderately positive and strongly positive,the number of both of which were 64,and the percentage of which were 62.14%.Phospho-NF-κBp65 protein expressed strongly in the nuclus of 84 ABs(81.55%in the total)sections,and its nuclear positive rate was 86.41%.The expressions of NF-κBp65 and Phospho-NF-κBp65 in ABs were obviously stronger than in normal mucosas,and there were statistics significance(X~2=38.904,P=0.000;X~2=34.755,P=0.000).There were close relationships between NF-κBp65 and Phospho-NF-κBp65,IκBα.NF-κBp65 had positive correlation with Phospho-NF-κBp65(r_s=0.383,P=0.000),negative correlation with IκBα(r_s=-0.645,P=0.000 ),and no relationship with IκBβ(r_s=-0.119, P=0.089).Furthermore,the difference of the four proteins in the primary group and recurring group has no significance(P>0.005).Conclusions The expression of NF-κBp65 protein in ABs was stronger than in normal mucosas,and it also expressed in some nuclears.Moreover,activated Phospho-NF-κBp65 protein highly expressed in the nuclear.lt was suggested that NF-κB promote the expression of its downstream factors when it translocated into nuclus.Some downstream factors possibly involved in the occurrence and development of the ABs,and enhanced the invasive ability of ABs.
