[objectives]Atopic dermatitis(AD) is a repeated pruritic skin disease, which might be accompained with serious complications. AD affects the life qualities of patients much, but utill now there is no satisfactory treatment,so it is the focus for all dermatologists long time.Atopic dermatitis is an inflammatory skin desease characterized by Th1/Th2 cell subtype imbanlance, and allergens in the enviroment play important roles in the ocurrence and development of AD.Combination tuckahoe soup is chosen based on the therapeutic principle of Tradtional Chinese Medicine(TCM) on eczema, which might affect Th2 and Th1 cell subtype proved by animal tests. In this article, anti-allergy and effects on the cytokin of TCM in Combination tuckahoe soup was further studied through the BALB/c mice model of atopic dermatitis induced by OVA, which help study pathogenesis of AD and find new TCM in the treatment of AD.[Methods] 170 healthy female BALB/c mice(body weight 25-30g) were randomly gruoped into 17 groups. Each group had 10 mice, which was cyproheptadine group, herba lammae group, fructus gardeniae group, rhizoma alismatis group, canker-root group, periostracum cicadae group, pachyma cocos Fr group, radices paeoniae rubra group, herba schizeneptae group, cortex phellodendri group, radices glycyrrhizae group, atractylodes sinensis group, radices sileris, radix angelicae sinensis group, flos lonicerae group, Normal Saline group, Hydrocortisone group respectively. One day before the test, Hair on mice abdomen was shaved, whose area was 3 mm2. Abdomen without hair was applied 20ml of 100mg/ml ovalbumin(OVA) once a day for consecutive 7 days. And every mice was fed TCM twice in the morning and in the evening a day. Then the mice were bred in the second and third week. On the forth week, the procedures of the first week were repeated, but 24 hours after the seventh day, the mice were killed, whose blood and skin lesions were taken. Serum IgE,IL-2,IL-4 and IFN-γlevel were tested with the adoption of ELISA. Skin lesions were prepared into HE sections and inflammatory cells were counted under microscopy.[Results]1.Compared with Normal Saline group, serum IgE level in flos lonicerae group decreased sharply, with obvious statistical difference (P≤0.01 or P≤0.001); serum IgE level in canker-root group, periostracum cicadae group, herba lammae group, radices paeoniae rubra group, radices sileris group deceased, with statistical difference(P<0.05).2.According to mice serum IgE level,canker-root, periostracum cicadae, pachyma cocos Fr, radices paeoniae, radices sileris and flos lonicerae were chosen to test IL-2, IL-4 and IFN-γlevel. Compared with Normal Saline group, serum IL-2 level in periostracum cicadae group, pachyma cocos Fr group, radices sileris group, flos lonicerae group increased, with statistical difference (P<0.05); IL-4 level in periostracum cicade group, pachyma cocos Fr group, radices paeoniae group, and flos lonicerae group decreased, with statistical difference(P<0.05). IFN-γlevel in pachyma cocos Fr group, radices paeoniae group and flos lonicerae group increased sharply, with obvious statistical difference(P≤0.01 or P≤0.001); IFN-γlevel in canker-root group and radix sileris group increased, with statistical difference(P<0.05).3.Compared with Normal Saline group, number of inflammatory cells of skin lesion in herba lammae group decreased sharply, with obvious statistical difference(P≤0.01 or P≤0.001); number of inflammatory cells in canker-root group, pachyma cocos Fr group, radices paeoniae group, radices sileris group and flos lonicerae group decreased, with statistical difference (P<0.05).[Discussion] Through the mice mode of atopic dermatitis,periostracum cicadae, radices sileris, canker-root, flos lonicerae and pachyma cocos Fr all can suppress serum IgE,on the basis of which, IL-2, Il-4 and IFN-γwere tested, with the results that periostracum cicadae, radices sileris, flos lonicerae and pachyma cocos Fr can up-regulate IL-2; periostracum cicadae, pachyma cocos Fr, radices paeoniae rubra, flos lonicerae and radices sileris can inhibit IL-4; canker -root, pahcyma cocos Fr, radices paeoniae rubra, flos lonicerae and radices sileris can upregulate IFN-γ, which might take effect by decreasing IgE level and regulating Th1/Th2 cells. IFN-γcan inhibit synthesis of more IgE by the inhibition of Th2 cells as well as decrease of IL-4 production, whose pathogenesis is expecting further study. In addition, herba lammae, canker-root, pachyma cocos Fr ,radices paeoniae rubra, flos lonicerae, radices sileris, radix angelicae sinensis may also inhibit inflammatory cells in skin lesions of mice, which might be associated with anti-inflammation and anti-allergy of TCM.
