Objective: (1) To study the expression of mda-7 on infant hemangioma endothelial cells;(2) To study mda-7 function on the apoptosis of infant hemangioma endothelial cells.Methods:(1) Infant hemangioma and normal vein tissues were acquired by surgical operation;(2) The location and protein level of mda-7 on infant hemangioma endothelial cells and normal vein endothelial cells were shown and measured by immunohistochemistry method and image analysis system;(3) The authors first obtained hemangioma and normal vein tissues. Total RNA was then extracted, then the expressions or levels of mda-7 in hemangioma and normal vein tissues were investigated by way of nesting polymerase chain reaction; (4) The protein level of mda-7 in hemangioma and normal vein tissues were shown and measured by western blot method.Results: (1)In normal vein tissues, the expression of mda-7 was weak or absent; In benign hemangioma tissues , it was stronger and it was the strongest in malignant hemangioma tissues. The expression location was located around nucleus membrane and in cytoplasm; (2)mda-7 mRNA was proved to be good not only in purity but also in intact by testing, average optical density were respectively 1.235±0.135, 0.101±0.089, There existed significant difference in these two groups (P<0.001); (3) Infant hemangioma group had stronger staining, indicating the expression of mda-7 protein was more, the staining was weak in normal vein group, there were significantly differences between relative coefficients of two groups(p<0.01).Conclusion:There was weak expression or no expression of mda-7 in normal vein tissues, the expression of mda-7 was positively relative to malignant extent in hemangioma tissues, embodying mda-7 might take part in the process of hemangioma occurrence,developing and apotosis by combining with its ligand.
