Objective:To investigate the expression level of WTX(Wilms Tumor gene on the X chromosome) gene in normal liver tissue,para-cancerous tissues and liver cancer tissues;To detect the expression of WTX gene in normal liver cells and hepatoma cells with different degree of malignancy; To analyze the relationship of WTX gene expression between normal liver cells and hepatoma cells in different degree of malignancy;To investigate the expression and significance of WTX gene in liver cancer.Methods:1、We collected 40 patient’s liver cancer tissues and para-cancerous tissue(from the edge of the lesion 2cm of the normal liver tissues) from the patient who don’t have other malignant tumor history, radiotherapy, chemotherapy and biological therapy before accepting operation treatment, setting up the hepatolithiasis and traumatic removal of part of normal liver tissue as a normal control group.2、The expression of WTX m RNA in the liver cancer tissues,normal tissues and para-cancerous tissues was detected by semiquantitative RT-PCR.3、The differential expression of WTX m RNA and protein in a normal liver cell line and different hepatoma cells line with different degree malignance were detected by RT-PCR and Western Blot,and was analysed the relationship of WTX between different hepatoma cells line.Results:1、The expression of WTX m RNA in hepatocellular carcinoma tissues with RT-PCR, the result suggested that cancer tissue was significantly lower than that of para-cancerous tissues and normal tissues, two the difference was statistically significant(P<0.05).2、The expression of WTX m RNA and protein in the normal liver cell was significant higher than those of liver cancer all lines, but there was no significant difference in other liver cancer cell lines(P>0.05).3、The expression of WTX m RNA and protein in the normal liver cell was significant higher than those of liver cancer all lines(P<0.05), but there was no significant difference in other liver cancer cell lines(P>0.05).Conclusions:The expression of tumor suppressor gene WTX was closely related to the malignant degree and tumor size in primary liver cancer tissue,and the WTX may have little relationship with the invasion and metastasis of hepatocellular carcinoma cell line.The deletion of the WTX gene expression may be one of the factors for the occurrence of primary hepatocellular carcinoma.
