Objective To investigate the effects of 2450MHz microwave irradiation on proliferation of cultured mouse fibroblast in vitro.And to analysis the influence of 2450MHz microwave on the expression of some genes and proteins related.Methods The cells from mouse skin were directly radiated by different intensity of microwave with different time.The growth metabolism of fibroblasts was assayed by MTT method,effects of microwave radiation on fibroblasts cell cycle were measured by flow cytometry.The mRNA expressions of the typeⅠandⅢprocollagen were detected by real-time RTPCR. Anti-phosphorylation extracellular-regulated Kinase(ERK-1/2) antibody were introduced in Immunofluorescence staining analysis to observe the phosphorylation changes of fibroblasts protein ERK.Results:①After radiation with microwave(0.5 W/cm~2,1.0W/cm~2,5W/cm~2) for 5min,15min,30min,fibroblasts activities reduced depending the radiated duration.After radiation with microwave(5W/cm~2) for more than 5min,fibroblasts activities reduced markedly(P<0.05).②After radiation with 5W/cm~2 for 5min,the percentage of cells in G0/G1 phase obviously was increased(P<0.05) and ERK were activated immediately after irradiation.③The mRNA expression of the typeⅠprocollagen was down-regulated after the HPM irradiation(P<0.05).The reduced expression levels were positively correlated with the irradiation duration.At 30min after irradiation with 1W/cm~2 and 5W/cm~2 for over 5min,the expression of typeⅠwas decreased significantly(P<0.05) and the expression of typeⅢprocollagen was't decreased significantly(P>0.05).But microwave irradiation downregulated the ratio of typeⅠandⅢprocollagen;After irradiation with microwave,the expression of MMP-9 and MMP-13 mRNA was increased significantly(P<0.05).④After radiation even of short duration using 2450MHz microwave,ERK were activated immediately after irradiation.Conclusion Fibroblast activities will reduce after radiation even of short duration using 2450MHz microwave in vitro.It is concluded that high-intensity microwave may inhibit the proliferation of fibroblasts in dose-dependent manner in vitro and down-regulate procollagen mRNA expression,which had correlation with decreasing of scar formation. Microwave irradiation can induced activation in MAPK signal conductive gateways.
