Objective To investigate change of gene expression spectra of heart in sepsis rats by DNA microarrays. Methods Thirty male Wistar rats were randomly divided into model group and blank control group with 15 rats in each group. Cecal ligationand puncture (CLP) was used to reproduce rat sepsis model and was identified by transmission electron microscope. Gene expression spectra was studied with oligonucleotide gene expression profile microarray that contains 22523 rat cDNA clones was used to detect the change in gene expression pattern of rat heart tissue 24 hours after CLP, genes with ratio average(RA)>2 or RA<0.5 were identified as variance genes,then differentially expressed genes that high correlated to sepsis were screened by means of related computer software, and the relationship between variance genes and sepsis was analyzed initially. Results Electron microscope demonstrated the modle fabrication succeeded. Compared to the controls, gene expression of 418 genes of hearts in rat sepsis model were chanced 24 hours after CLP, accounting for 1.86%,among them 200 genes expression up-regulated, 218 genes expression down-regulated. Among the known unique genes, 84 of them were up-regulated,included 25 acute stress reaction related genes,10 signal transduction related genes,4 transcription factor related genes,6 immune response related genes,4 ion channel related genes, 7 energy metabolism related genes, 2 DNA duplication related genes, 2 growth factor related genes, 2 cancer related genes,2 inflammatory response related genes, 1 angiotensinogen and 1 angiotensinI converting enzyme related genes,15 for the rest; 74 of them were down-regulated,included 16 energy metabolism related genes, 10 signal transduction related genes, 6 inflammatory response related genes,3 redox regulation related genes, 5 cytoskeleton related genes, 4 acute stress reaction related genes, 6 immune response related genes, 5 growth factor related genes, 2 transcription factor related genes, 1 ion channel related genes, 1 DNA repairing related genes, 15 for the rest. Conclusion①The significantly up-regulated genes included gene AGT and ACE.②Genes related to G-protein mediated signal transduction and CEBPB were up-regulated .③Gene related to glucocorticoid receptor was down-regulated .④Genes related to nonspecific immunity were up-regulated , Genes related to specific immunity were down-regulated .⑤Genes related to insulin-like growth factor were down-regulated.⑥A majority of the energy metabolism related genes were down-regulated.⑦Further study using the technique of DNA microarrays may yield great insight into the molecular pathologic mechanism of sepsis and discern new targets for therapeutic interventions.
