Objective:To identify and analyze differential expression of UCHL1 in rat offspring testes after maternal exposure to di(nbutyl)phthalate (DBP).Methods:Forty pregnant rats were randomly divided into two groups and were given DBP by gastric intubation at a dose of 800 mg/kg·d or none from the 14th day to the 18th day of pregnancy.Testes form normal or exposure group were taken out respectively at GD19 and PND22.we use western blotting and Immunohistochemistry to detect and analyze the expression of UCHL1.Results:UCHL1 expression in DBP-exposed group was significantly 50% lower than the normal controls at GD19.but at PND22,the two groups are almost equal.Immunohistochemical analyses showed that UCHL1 expressed higher in cell nucleus and cytoplasm of spermatogonias of normal fetal rat testis than DBP exposure group at GD19. Conclusion:The finding suggested that The decrease of UCHL1 after DBP in utero exposure and disturbance of ubiquitin-proteasome system may be responsible for the maldevelopment of spermatogonias.
