The Expression And Clinical Significance Of Caspase-3 And Caspase-6 Proteins In RCCC

OBJECTIVE: 1 To investigate the expression of Caspase-3 and Caspase-6 proteins in human renal clear cell carcinoma (RCCC),para-tumor tissue and normal renal tissue.2 To probe into the expression of Caspase-3 and Caspase-6 proteins were correlated with the clinical pathologica characters and prognosis of RCCC.3 To analyze the relation of Caspase-3 and Caspase-6 proteins in RCCC.METHODS: 75 RCCC and their para-tumor tissue and 15 normal renal tissue were analyzed for expression of Caspase-3 and Caspase-6 proteins by SP immunohischemistry and correlation with the characters and prognosis of RCCC.RESULTS:1,The positive rate of Caspase-3 protein in all the RCCC patients was 74.67% , significantly lower than that in para-tumor tissue (92.00%) (P<0.01). In G₁, G₂, G₃ of RCCC, the positive rate of Caspase-3 protein were 87.50%, 75.68%, 53.85%, the diference only between G₁ and G₃ (P <0.05) .In I, II , III～IV of clinical stage , the positive rate of Caspase-3 protein were 87.97%, 68.18%, 63.64%, the diference only between I and III～IV (P<0.05). The positive rate and the negative rate of Caspase-3 protein in that with lymph nodes metastasis were 55.56% and 80.70%,the diference between them was significantly(P<0.05).2,The positive rate of Caspase-6 protein in all the RCCC patients was 76.00% , lower than that in para-tumor tissue (90.67%) (P<0.05). In G1, G2, G3 of RCCC ,the positive rate of Caspase-6 protein were 83.33%, 76.32%, 61.54% and in I,II, III～IV of clinical stage the positive rate of Caspase-6 protein were 80.65%, 68.18%, 77.27%,the diference among them was not significant (P>0.05) . The positive rate and the negative rate of Caspase-6 protein in that with lymph nodes metastasis were 75.44%, 77.78% ,the diference between them was not significant (P> 0.05 ) .3,In RCCC tissue ,the negative expression of Caspase-6 protein were 12 cases in the positive expression of Caspase-3 protein ; the negative expression of Caspase-3 protein were 13 cases in the positive expression of Caspase-6; the positive expression of Caspase-3 and Caspase-6 proteins were 44 cases; the negative expression of Caspase-3 and Caspase-6 proteins were 6 cases. The expression of Caspase-3 and Caspase-6 proteins may not correlated with each other .4,After 3 years follow-up in 35 cases,the mortality rate. in 3 years of the positive and negative expression of Caspase-3 protein were 36.00% and 40.00 %;that of Caspase-6 protein were 34.78% and 41.67%;the mortality rate in 3 years of the positive expression Caspase-3 and Caspase-6 proteins was 30.00%; the negative in all was 66.67%. The diference between them each other was not significant (C=0.0921 ,P > 0.05) . CONCLUSIONS:1,The positive expression of Caspase-3 protein in RCCC was significantly lower than that in para-tumor tissue and normal renal tissue. It may be correlatied with the clinical stage and pathologic grade of RCCC and with lymph nodes metastasis in RCCC.2,The positive expression of Caspase-6 protein in RCCC was significantly lower than that in para-tumor tissue and normal renal tissue. It may not be correlatied with the clinical stage and pathologic grade of RCCC and with lymph nodes metastasis in RCCC.3,In RCCC, the lower positive expressions of Caspase-3 and Caspase-6 proteins were independence from each other ,but may be both correlated with the apoptosis of the cancerous cells. It may elevate the curative effect of RCCC to promote their expressions targetedly. The relationship of the prognosis of RCCC with the lower expression of Caspase-3 and Caspase-6 proteins is not clear.
